Objiect: The relation between uric acid and tissue plasminogen inhibitor-1(PAI-1) remains unclear. Therefore, this study was aimed to examine the effect of uric acid on the production of PAI-1 in HUVECs and to identify the putative underlying signaling pathways. Methods: Cultured HUVECs were stimulated with uric acid; cell proliferation and PAI-1 proteinum expression were examined. The effect of uric acid on extracellular signal-regulated kinases (ERK) phosphorylation were tested to elucidate the intracellular mechanism of uric acid in PAI-1 proteinum expression. Results: Uric acid inhibed the HUVECs proliferation and increased PAI-1 proteinum expression. Uric acid also increased ERK phosphorylation,. PD98059 suppressed partially uric acid-induced PAI-1 proteinum expression.,.Conclusions: These results suggest that uric acid-induced PAI-1 proteinum expression, partially by the activation of ERK pathway in HUVECs.
