Objective:1.To establish a rabbit model for intractable constipation;2.To explore matrix metalloproteinase -2 (MMP-2) and typeⅣcollagen in colon of constipated rabbits,and research on the relationship between them with intractable constipation;3.We put a pipe into rabbit's colon and wash,observe and summarize effects of treatment,it may be provide a basis theory for clinical practice in the future.Methods:Thirty female rabbits were randomly divided into two groups.There were fifteencases in each group. Fifteen cases in experimental group were used to establish the model of intractable constipation. A tooting was fixed on anterior wall of rectum in experimental rabbits .Fifteen cases in control group were bred normally with forage. Two months later all rabbits'colon were visualization by clyster of X line barium. We put pipes into experimental group's colon, obtain a small tissue of colon and measure all rabbits length of colorectum and diameter . Then we analyzed these data to compare if there was any obvious statistical significance between two groups.We assayed MMP-2 and typeⅣcollagen of all rabbits'colon by SABC immunohistochemical method. We analyzed images and data by Image-Pro Plus 6.0 and SPSS13.0.Results:We established rabbit model for intractable constipation successfully. Barium enema showed that rabbits in experiment group colon was tediously long and winding.The overall length of colorectum became longer and the diameter of colon became thicker, The content of experiment group colon MMP-2 was significantly higher than control group (D 83.37±13.10vs41.04±16.44 P<0.01), and the content of typeⅣcollagen was significantly lower(D 93.41±19.74vs187.85±22.79 P<0.01). Experimental rabbits had thin and soft feces by washing, effective power of treatment was 100% , the overall length of colorectum became shorter and the diameter of colon became thinner.Conclusion:1.We established successfully animal model of intractable constipation disease;2. Increase of MMP-2 of rabbits'colon tissue and degradation of typeⅣcollagen,they promoted each other with intractable constipation, it was the very important cause and factor for this disease;3. Treatment of putting a pipe into colon and washing became effective dramaticlly in short term,it was a convenient and quick treatment for intractable constipation. it provided the basic treatment of theory for clinical implementation in the future.
