objective: To explore the application of MRI(Magnetic resonance imaging) on the transplantation of MSCs(Marrow mesenchymal stem cells) to the hippocampus And to examine the effect of the transplantation on PSA-NCAM in diabetic rat.Methods: 30 SD male rats were divided into the normal control group,diabetic model group, transplant group randomly. Streptozotocin-diabetic rat model was established. The learning and memory ability of rats were tested with the Morris water maze after 12 weeks. Primitive MSCs were cultured and SPIO was used to labeling the MSCs before they were transplanted to the right side of the hippocampus of the diabetic model group with the other side left to be contrast. The images of the first day, the third day, the seventh day and the nineth day were collected by MR to observe the planting of MSCs. Then compare the protein level of PSA-NCAM in hippocampus by immunofluorescence and the expressions of PSA-NCAM mRNA in hippocampus by RT-PCR among three groups.Results: 1.Compared with diabetic model group,the normal control group showed a significant decrease in the mean time of escape latencies and a increase in percentage of stay time in the central area and the number of passing the original platform position.2.MR scanning shows there are signal reduce blots in the left part of hippocampus with T2WI/FSE,T2*WI/GRE sequences,especially in GRE signal.3.Compared with diabetic model group,the expression of protein PSA-NCAM in hippocampus of transplanted group showed a significant increase(P < 0.05 ) ,while compared with control group, the protein PSA-NCAM showed a significant decrease(P<0.05).4.Compared with diabetic model group,the expression of PSA-NCAM mRNA in hippocampus of transplanted group showed a significant increase(P<0.05),while compared with control group, the PSA-NCAM mRNA showed a significant decrease(P<0.05).Conclusion: 1.DM rats have disability in learning skill and damage in memory 12 weeks afer they have modellized.2.The MRI of the 1st,3th,7th,9 th showed labeled MSCs can imaging stably in hippocampus of DM rats,and they will not disappear with time.3.The change in expression of PSA-NCAM in hippocampus of rats showed neuron differentiation in DM rats after MSCs transplanted.
