Objective: Making a model of PCOS rats, we can compare the ERα,ERβexpression of PCOS rats and the normal ones, in their endometrium, also in cervix. To determine the expression of ERα,ERβin the endometrium and cervix of PCOS rats.Method: 50 female SD rats of 42 days, were randomly divided into 2 groups:①the control group (20 rats),which were injected subcutaneously with 0.2ml sesame oil for 28 days.②the model group (30 rats): the dehydroepiandrosterone (DHEA) and insulin (Ins)were used to construct the PCOS rats for 28 days .When the rats were at 70 days, were observed changes in estrous cycle by consecutive vaginal smears for 20 days, While at 90 days, 20 of model rats were randomly selected (the left 10 were consecutive vaginal smears till to 110 days, to observe the stability of the model) to determine the level of T, E2, P, LH, FSH, FPG, FINS in the serum, and their ovaries were excised to be given ovarian histological detection, and so did the 20 control rats in the same period. Immunohistochemistry, RT-PCR were used to detect the expression level of ERα, ERβprotein and mRNA in endometrium, also in cervix tissue of the two groups .Results:①The model group lost the regular estrous cycle, and the other 10 model rats , used vaginal smear 40d(70d ~ 110d) for observating the stability ,did not restore the regular estrous cycle.②the model group rats'ovaries seemed to be polycystic change, follicle saccular ectasia, granular cell layer decreased, and corpora atretica were seen.③the level of T,LH,LH / FSH,FINS,FPG in serum of the model group were significantly higher than the control group (P<0.01), P levels were significantly lower than the control group (P<0.01), E2,FSH had no significant difference (P>0.05) between the two groups.④Immunohistochemistry showed that The expression of protein of ERαin PCOS group in endometrium was significantly higher then those in the control group (P<0.01);whereas ,the expression of protein of ERβin endometrium was not significantly different between the two groups(P>0.05). RT-PCR analysis showed the same expression of ERαmRNA and ERβmRNA in endometrium as immunohistochemistry.⑤Immunohistoch- emistry showed that The expression of protein of ERαin PCOS group in cervix significantly higher then those in the control group(P<0.01); whereas ,the expression of protein of ERβin cervix was not significantly different between the two groups(P>0.05). RT-PCR analysis showed the same expression of ERαmRNA and ERβmRNA in cervix as immunohistochemistry. Conclusion:①ERαin endometrium of PCOS rats is expressed excessively.②ERαin cervix of PCOS rats is expressed excessively.
