Objective:To investigate the expression of the hTERC oncogene in cervical scraping samples,and to evaluate the effect and clinical significan- ce of hTERC in cervical lesion.Atypical dysplasia cervical cells transforms to the cervical cancer almost all associated with the long arm of chromosome 3 amplification.The RNA component of the human telomerase gene (TERC) on chromosome band 3q26. may be play the the most important role in these.A majority researches indicate that chromosome instability is the important process in the Cervical cancer effluence.The chief manifestations are the abnormality of number or structure in chromosome and the amplification or deletion of DNA series and the change of base,etc.Fluorescence in situ hybridization is a technology of molecular genetic developed in recent ten years.Its principle is displaying the position of DNA series in nuclear or chromosome through nucleic acid probe with fluorescent material.It can detect the DNA series and its diversity in the metaphase and interval cell.The technology is helpful for early diagnosis of Cervical cancer in detection of the rhagiocrine cell of Cervica1 intraepithelial neoplasia,it also helps evaluate the criterion of prognosis or relapse of Cervical cancer.Methods:The fluorescence signal of cytologic samples of cervix were detected by using interphase FISH in chromosome enumeration doublecolor DNA probes TERC.According to histology biopsy,130Pap smears divided into normal(n=25),cervical intraepithelial neoplasia(CIN,n=78),squamous carcinomas of the cervix(SCC,n=27).Results:None normal samples revealed copy number increases of 3q,while 18.75% of the CINI,62.9% of the CINII/III lesions and 92.59% of the squamous cervical cancer showed extra copies of 3q.TERC gene copy numbers in CINII/III were significantly higher than that in CINI.The percentage of multiple 3q signals increased with the severity of the cytologic interpretation(P<0.05).Conclusion:The increase of hTERC expression in the invasive SCC and CIN suggests that 3q copy numbers are associated with the severity of cytologic and histologic findings.hTERC gene of cervical scraping sam- ples is an early event and could be used as a better tumor index for early cervical cancer screening and predicate the the advancement cervical lesion.
