Objective:To analyze the uptake of 99Tcm–HL91,expression of HIF-1α,the change and regular pattern of apoptosis cells in breast carcinoma tissues after the intervention of hyperbaric oxygenation,to inve- stigate the contribution of hyperbaric oxygenation to the uptake of 99Tcm–HL91 and cells apoptosis.Methods: 40 breast carcinoma761 mice,pantheress 615,are randomized 4 groups,each 10mice:control group were performed 99Tcm-HL91 rad- ionuclide imaging only;The hyperbaric oxygen group were performed hyperbaric oxygen for 1h after injecting 99Tcm-HL91 and radionuclide imaging after 4h; The chemotherapy group were performed 99Tcm-HL91 radionuclide imaging after injection Paclitaxel in peritoneal cavity by 5mg/kg after 24h;The hyperbaric oxygen chemotherapy group were treated in HBO for 1h after injection Paclitaxel and were performed 99Tcm-HL91 radionuclide imaging after 24hours of injection.Every group were performed whole body imaging after 4h injection about 37MBq 99Tcm-HL91 by caudal vein.Mice were executed immediately for dipping and immo- bilization of specimen after acquisition.It was measured the amount of apoptosis cells with TUNEL and HIF-1αwith immunohistochemisty. The results were calculated the ratio of tumor and the contralateral corresponding site with ROI.Results: All of the tumors display dense uptake of nuclide and its margin outstand of the normal. The positive cells by TUNEL can be seen in every section.The positive results by HIF-1αcan be seen in every section. The ratio of T/NT,amouts of apoptosis cells and the value of HIF-1αis 2.309±0.306,4.50±0.926,0.010±0.004 in control group,1.964±0.194, 5.20±1.033,0.005±0.002 in hyperbaric oxygen group,2.017±0.194, 8.90±2.132,0.005±0.001 in chemotherapy group,1.690±0.205,12.30±2.584,0.004±0.001 in hyperbaric oxygen chemotherapy group. The result of T/NT has significant difference (t=-2.489,P=0.042),and so it is optical density (t=-2.347,P=0.017),but the result of apoptosis cells amout has no significant difference (t=-1.594,P>0.05).The differences of T/NT,the amount of apoptosis cells and the means value of optical density were of significance in statistics science between hyperbaric oxygen group and chemotherapy group (t=-2.489,-3.098,-3.111;P<0.05).The value of T/NT and optical density had positive correlation in all groups (r=0.856, 0.842,0.811,0.728;P<0.05),The amount of apoptosis cells and optical density had negative correlation (r=-0.807,-0.681,-0.916,0.880;P <0.05). The amount of apoptosis cells and T/NT had negative correlation in chemotherapy group and hyperbaric oxygen chemotherapy group (r=0.682,-0.907;P<0.05).Conclusions:HBO can decrease the uptake of 99Tcm-HL91 and expression of HIF-1αin breast carcinoma mice; HBO can enhance the introduction of Paclitaxel to apoptosis in breast carcinoma mice; 99 Tc m -HL91 imaging and HIF-1αexpression have the consistency in detection of anoxia in breast carcinoma mice .
