Objective:This experiment is designed to set up animal models of duck hepatitis B was observed at the ganluxiaodudan addition and subtraction of duck hepatitis B virus in vivo whether inhibition and nucleoside antiviral drug lamivudine compared to,to explore its mechanism.Methods:(1)Manufacture of animal models:Screened using PCR method D-HBV positive ferruginea extracted D-HBV positive serum,with leg venous inoculation three-day-old duck, set up the day after tomorrow duck hepatitis B virus infection in animal models.(2) the experimental observation:the D-HBV positive ferruginea 40 were randomly divided into 5 group,model control group(A group),the ganluxiaodudan addition and subtraction high dose group(B group),the ganluxiaodudan addition and subtraction middle dose group(C group), add the ganluxiaodudan addition and subtraction low-dose group(D group) and lamivudine group(E group);the group given conventional feed,drinking water freely.The observation group were given different doses of the ganluxiaodudan addition and subtraction(B group: 30g·Kg.-1d-1;C Group:20g·Kg.-1d-1;D Group:10g·Kg.-1d-1) and Pascal Lamy stavudine (50mg·Kg.-1d-1) to intervene.Pre-treatment,treatment of 7,14,21 days and seven days after discontinuation of treatment,respectively,blood samples 0.2ml,to 3000 per minute centrifugal extraction of serum,-20℃refrigerator to preserve the seizure preparedness. Sacrificed before treatment D-HBV-positive ducklings 2,filling the first 21 days post-drug the ganluxiaodudan addition and subtraction high-dose group and lamivudine group two of the ducks,take liver tissue Add 4%formaldehyde fixation conventional HE staining inflammation.(3) laboratory tests:Using real-time fluorescence quantitative PCR(SYBR Greenâ… ) dynamic testing before treatment,drug intervention in the first 7 days,14 days,21 days,seven days after stopping D-HBV serum concentrations and serum enzyme Alarelin acid aminotransferase(ALT),aspartate aminotransferase(AST) Change;in the optical microscope(×400 times) before and after treatment in each group to observe the pathological changes in liver.Results:Lamivudine group after administration to quickly lower the serum virus levels and treatment has been to maintain a lower level,the ganluxiaodudan addition and subtraction of high,medium dose group,respectively,serum viral load drop at the first day of 14,21 markedly(with the model control group,inter-group comparison p<(0.05),low-dose group during treatment not inhibit the role of virus.After drug the ganluxiaodudan addition and subtraction high,medium-dose group and model control group still demonstrated a certain degree of inhibition of virus;and lamivudine serum virus titer rebounded.ALT,AST before and after treatment compared to lamivudine group after treatment ALT,AST rapidly reduced, the treatment has been to maintain the normal level,the ganluxiaodudan addition and subtraction of high,medium dose group ALT,AST,respectively,beginning at the first days 14,21 markedly(with the model control group,inter-group comparison p<(0.05),low-dose group had no obvious role during treatment.After drug the ganluxiaodudan addition and subtraction of high,medium dose group ALT,AST continued rehabilitation regular status. And lamivudine group ALT,AST increased again.Liver tissue biopsy after treatment showed that addition and subtraction Dan Ganlu disinfection high-dose group and lamivudine group to improve the situation quite inflammatory.Conclusions:the ganluxiaodudan addition and subtraction indeed D-HBV-induced hepatitis have therapeutic effects,it can not only inhibit virus replication in vivo,but can also reduce the immune response so that ALT,AST normalization and reduce liver inflammation.
