Objective: To study the effects of Macleya cordata B on hepatic pathology,hydroxyproline level,cytokines expression and serum markers of fibrosis in rats in the development of hepatic fibrosis,and to discuss the mechanism of Macleya cordata B in the treatment of hepatic fibrosis.Methods:Models of hepatic fibrosis were established by hypodermic injection of tetrachloride, in combination with the control of nutrition and the drinking of 10% alcohol to rats. Five rats were killed at each weekend to examine pathological changes. According to histological sections, hepatic fibrosis in rats emerged at the end of the fourth week. The rest 75 rats could be used for the next study which were then divided into five groups as Fibrotic model group(group B), Interferon-gamma treatment group (10MU/kg.d,im) (group C), Low-dose Macleya cordata B group (0.2mg/kg/d,i.p.) (group D), Medium-dose Macleya cordata B group (0.6mg/kg.d,i.p.) (group E) and High-dose Macleya cordata B group (2mg/kg.d,i.p.) (group F). In addition, another 10 rats were chosen to be the normal control group(group A),Rats in the Group A and Group B were given respectively 0.9% sodium chloride by peritioneal injection instead of Macleya cordata B. Rats in other groups received 8-week's Macleya cordata B or Interferon-gamma treatment. At the end of the eighth week, all the rats were executed. Histological sections of liver from the killed animals were evaluated by hematoxylin and eosin, Van Gieson and Masson stains. Transfoming growth factor-β1(TGF-β1) and a-smooth muscle actin (a-SMA) were examined with immunohistochemistrical technique. What's more, Liver hydroxyproline content and serum hyaluronic acid level,Laminin,PCⅢ,Ⅳ.C were also detected.Result:1The pathological fibrosis scores, the expression of TGF-β1 and a-SMA in live , liver hydroxyproline content ,serum levels of HA, LN, PCIII,ⅣC of rats in fibrotic model group induced by CCL4 increased significantly compared with normal group(P<0. 01 ),which were ameliorated in group C to F (P<0. 01 ).2 The change of the pathological fibrosis scores, the expression of TGF-β1 and a-SMA in live , liver hydroxyproline content ,serum levels of HA, LN, PCIII,ⅣC was not obviously different between all Macleya cordata B groups andγ-INF group. 3The pathological fibrosis scores, the expression of TGF-β1 and a-SMA in live , liver hydroxyproline content decreased significantly in middle-dose group compared with low-dose group(P<0.05), which was not obvious distinguish from high-dose group(P >0. 05).Conclusions:1 Macleya cordata B can improve hepatic pathology,reduce liver hydroxyproline content and the expression of TGF-β1 as well as a-SMA in the development of liver fibrosis in rats. Macleya cordata B also has the effect of lowering serum markers of fibrosis.2 The anti-fibrotic effects of Macleya cordata B are identical with IFN-γ.3 Macleya cordata B has certain dosage-effect scope in treating hepatic fibrosis rats.
