Objective: S-adenosyl-L-homocysteine(AdoHcy) is a histone methylation inhibitor, Trichostatin A(TSA) is one of histone deacetylation inhibitors. To research the role of Adohcy and / or TSA for the gastric cancer SGC-7901 cells, to explore their effection for the expression of the tumor-related genes P21WAF1 in gastric cancer cells, and the possible mechanism of their inhibition on gastric cancer cell proliferation .Methods : Conventional cultured human gastric cancer cell line SGC-7901 in vitro, cells were treated with AdoHcy,AdoHcy+TSA at different time points and concentrations.MTT assayed the inhibition role of AdoHcy,AdoHcy+TSA for the growth of SGC-7901 cells, Cell cycle was determined by flow cytomery, RT-PCR and Western-blotting detected mRNA and protein expression changes of P21WAF1 gene.Results : 1.The experimental results of MTT show that: After treating with different concentrations (0.1, 0.2, 0.4, 0.8 mmol/L) AdoHcy for 24,48,72h on the gastric cancer SGC-7901 cells, AdoHcy could inhibit the growth of gastric cancer SGC-7901 cells in a time and dose dependent manner; comparing different concentrations (0.1, 0.2, 0.4, 0.8 mmol/L) AdoHcy +400ng/mlTSA with the same concentration of AdoHcy group, the inhibition rate of combination group increased significantly, performance for the combined or synergies (P <0.01).2. Flow cytometry analysis found that treating with different concentrations AdoHcy (0.1, 0.4, 0.8 mmol / L) for 48h on the gastric cancer SGC-7901 cells , they induced cell cycle arrest,the cells were arrested in G1 phase.With the increase of concentration, the percentage of cells in G1 phase increased, accompanied by a reduced proportion of cells in S phase, G2 index without significant changes; comparing different concentrations (0.1, 0.4, 0.8 mmol / L)AdoHcy +400ng/mlTSA with the same concentration of AdoHcy group, inhibition of combination group was significantly increased (P <0.05).3. AdoHcy and AdoHcy + TSA induced the expression of P21WAF1 in a dose dependent manner in SGC-7901 cell , comparing with single drug group,the combined group increased the expression of P21WAF1 significantly (P <0.05). Conclusion:1. AdoHcy and AdoHcy + TSA could inhibit the cell proliferation of gastric cancer SCG-7901 cells in a time and dose dependent manner, induced cell cycle arrest, the cancer cells arrest in the G1 phase, and increased the expression of P21WAF1 gene in a dose dependent manner . Comparing with single drug group,the combined group could significantly inhibit the cell proliferation and promote the expression of P21WAF1 in gastric cancer SCG-7901 cells.2.The inhibition of AdoHcy and AdoHcy + TSA in gastric cancer SCG-7901 cells may relate with G1 phase arrest, increasing the expression of P 21WAF1.
