Burdock is a large-scale biennial herbs compositae. Its own a variety of secondary metabolites. This study from the perspective of resource conservation, external goods used as raw materials extraction to study the burdock the extraction of inulin.The three methods of hot water, ultrasonic and microwave to extract inulin from the Arctium lappa L. were studied in this paper. The factors that influenced the extraction, such as ratio of solid to liquid, temperature, time and ultrasonic power were investigated and the optimum ultrasonic extraction conditions were obtained through the orthogonal experimental design. The results showed that the method of ultrasonic can extract inulin effectively. The extraction yield rate was 49.31 under the optimum extracting conditions: ratio of solid to liquid 1: 13,time 45min,temperature 65℃, ultrasonic power 90W.The study also discussed the purification of inulin. The inulin solution was de-protein by Seveage method, precipated by ethanol purified burdock oligosaccharide was gained. The purity of inulin with purity was about 91 %.To establish the osteoporosis model of castrated rat and use the model to study the effect of inulin on the absorption and utilization of calcium in rats. The results showed that the calcium absorptivity of middle and high dose inulin group were higher than the sham-operation group by 15.15% and 19.6%.The antioxidative activities, including hydroxyl radicals and 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radicals were studied. The results indicated that the extraction of burdock exhibited high free radical scavenging activity, and their reducing power was equivalent with Ascorbic Acid.Through tahen the extraction of burdock, the activities of the antioxidant enzymes SOD, GSH-Px, TAOC were higer than control group(p<0.05), and MDA was lower than control group(p<0.05) in blood serum. The activities of the antioxidant enzymes SOD, GSH-Px, TAOC were higer than control group(p<0.05) in liver tissue. The activities of the antioxidant enzymes SOD, GSH-Px, TAOC were higer than control group(p<0.05), and MDA was lower than control group(p<0.05) in brain tissue. Through tahen the extraction of burdock, the activities of the antioxidant enzymes SOD mRNA had a remarkbale gene expression level(p<0.05). Otherwise, the extraction of burdock can protect liver tissue structure, help relieve the reduce of nerve cell, and also can improve the symptom of interstitial nephritis.
