| IntroductionOvarian cancer is one of three malignant tumors in the female reproductive system, serous cystadenocarcinoma is the most common malignant ovarian tumor in ovarian cancer,accounting for 40-60%of malignant ovarian tumors,therefore,it is important to explore effective diagnosis and treatment of ovarian serous cystadenocarcinoma.Integrin is widely a class of cell adhesion molecules in animal and plant cell surface,heterologous catamaran transmembrane glycoprotein composed ofαandβsubunits by non-covalent bond.α5β1 is composed ofα5 andβ1 subunit.α-subunit is related to identify the ligand,βsubunit transduct signal to the cytoskeleton.In recent years,studies have shown there is a certain relationship between integrinα5β1 and the occurrence and the development of malignant tumors.It affect directly or indirectly the transfer and apoptosis of tumor cells,promote generation of tumor vascular by many links such as adhesion and signal transduction between tumor and the host cell, basement membrane,thereby promoting tumor cell growth and metastasis.Glycosyl antigen is also expressed in the cell membrane.It is glycoconjugates composed of the sugars and proteins or lipids by covalent binding,involved in many cell functions,Including adhesion,recognition and signal transduction.Lewis y antigen is the end of oligosaccharides in glycoprotein and glycolipid,is typeⅡblood group-associated antigen.Lewis y was found in epithelial tissues,including ovary, thymus,lung,stomach and colon,etc.When these organizations occurs canceration, expression was significantly increased,and the patients increased Lewis y was with poor prognosis.In this study,we selected cases of paraffin-embedded including ovarian serous cystadenocarcinoma,ovarian borderline serous cystadenoma,ovarian serous cystadenoma,and normal ovarian specimens.Using immunohistochemical staining methods,detect the expression of Lewis y antigen,α5 andβ1 integrin.Discussion the the mutual relations between Lewis y antigen,integrinα5 andβ1 and ovarian serous tumors clinical pathology.Methods1.SubjectSelect 64 cases of paraffin-embedded specimens from 2006 to 2008 in China Medical University Hospital Obstetrics and Gynecology Shengjing,all casea have been confirmed by pathology experts after HE staining,all cases were Divided into four groups,Ovarian serous cystadenocarcinoma were 30 cases,patients age were 36～73 years old,the average age was 55.7 years old;Borderline ovarian serous cystadenoma group 10 cases,patients age were 22～55 years old,the average age was 33.8 years old; ovarian serous cystadenoma group 13 cases,patients age were 15～72 years old,the average age was 44 years old;Normal ovarian group 11 cases,patients age were 37 52 years old,the average age was 43.6 years old.2.Immunohistochemical detection of Lewis y antigen,integrinα5 andβ1Deparaffinize in xylenes using three changes for 5 minutes each.Hydrate sections gradually through graded alcohols:wash in 100%ethanol twice for 10 minutes each, then 95%ethanol twice for 10 minutes each.Wash in deionized H2O for 1 minute with stirring.Aspirate excess liquid from slides.Incubate specimens with 10%normal blocking serum in PBS for 20 minutes to suppress non-specific binding of IgG. Blocking serum ideally should be derived from the same species in which the secondary antibody is raised.Wash with PBS.Incubate with primary antibody for 60 minutes.Optimal antibody concentration should be determined by titration; recommended range is 0.5-5.0μg/ml in PBS with 1.5%normal blocking serum.Wash with three changes of PBS for 5 minutes each.Incubate for 45 minutes with secondary antibody diluted to 1-5μg/ml in PBS with 1.5%-3%normal blocking serum.Optimal antibody concentration should be determined by titration.Wash with three changes of PBS.Wash extensively with PBS.Mount coverslip with aqueous mounting medium or 90%glycerol in PBS.Results1.The expression of integrinα5. The expression of integrinα5 was 86.7%(26/30) in ovarian serous cystadenocarcinoma group,72.7%(8/11) in borderline ovarian serous cystadenoma group,69.2%(9/13) in ovarian serous cystadenoma group,36.4%(4/11) in normal ovarian group.2.The expression of integrinβ1.The expression of integrinβ1 was 83.3%(22/30) in ovarian serous cystadenocarcinoma group,72.7%(8/11) in borderline ovarian serous cystadenoma group,53.8%(7/13) in ovarian serous cystadenoma group,36.4%(4/11) in normal ovarian group.3.The expression of Lewis y antigen.The expression of Lewis y antigen was 90.0%(27/30) in ovarian serous cystadenocarcinoma group,63.6%(7/11) in borderline ovarian serous cystadenoma group,38.5%(5/13) in ovarian serous cystadenoma group,0%(0/11) in normal ovarian group.4.Relations between Lewis y antigen,integrinα5 andβ1There is a positive correlation between the expression of Lewis y antigen and integrinα5.there is also between the expression Lewis y antigen and integrinβ1.ConclusionThe expression of Lewis y antigen,integrinα5 andβ1 increased in Normal ovarian group,ovarian serous cystadenoma group,Borderline ovarian serous cystadenoma group and Ovarian serous cystadenocarcinoma group,there is a positive correlation between the expression of Lewis y antigen and integrinα5,Lewis y antigen and integrinβ1.
