IntroductionUlcerative colitis(UC) is a kind of chronic nonspecific intestinal inflammation without definite cause.The pathogenesis and etiology are so complicated that they are not very clear by now.Many recent research show UC have relation with blood hypercoagulable state,microthrombus formation may be one pathogenesis of UC, hypercoagulable persistently state may have relation with clinical progression of UC, which can promote inflammation take place and progress.Pathological process and clinical complication in the illness with thrombus formation have close relations with tissue factor(TF) and tissue factor pathway inhibitor(TFPI).Recent reach show TF and TFPI have relations with inflammatory reaction too,TF and TFPI can express in many kinds of tissue and cells.There are few reports about the relations between TF and TFPI and UC by now.In this study,we performed immunohistochemical staining for TF and TFPI expression in tissue of UC and normal colon,find out the relations between which and UC and reveal their significances in pathogenesis.MaterialThirty pathological specimens of UC tissue from the patient with UC in active stage under the colonscope and thirty pathological specimens of normal tissue from the patient without UC under the colonscope diagnosed by pathological doctor after HE stained from pathological department of Fengtian hospital affiliated to Shenyang Medical College are seleced.The study cohort consisted of 28 men and 32 women (mean age 49 years,range 35-52years).Proof of the UC diagnosis:typical clinical manifestation and specific changes under the colonscope.Main reagents:Anti-Human TF antibody,Anti-Human TFPI antibody, UltraSensitiveTM SP,KitS Reagent boxMain instruments:Microscope(OLYMPUS BX40),refrigerator(rongsheng)MethodMethod:expressions of TF and TFPI in UC are detected by SP immunohistochemical stain.Statistic Analysis:SPSS11.0 statistic software was applied.All data were analyzed withχ2 test and p<0.05 was regarded as statistically significance.Results1.Expression of TF in tissue of UC and normal colonThe positive rate of TF in tissue of UC is 63%,and the positive rate of TF in tissue of normal colon is 33%.There is significant difference between the two groups compared(p<0.05).2.Expression of TFPI in tissue of UC and normal colonThe positive rate of TFPI in tissue of UC is 43%,and the positive rate of TFPI in tissue of normal colon is 17%.There is significant difference between the two groups compared(p<0.05).DiscussionClinical observation show patient with UC have blood hypercoagulable state and potential danger of thrombus formation,hypercoagulable persistently state may have relation with clinical progression of UC,which can promote inflammation take place and progress.It is definite that extracellular part of TF connected with FⅦcan start exogenous coagulate pathway,which have thromboplastic activity and proteolysis.TF have significant in physiological coagulation and pathological thrombus formation,and participate in inflammatory reaction.Phosphorylation of three residues of serine of intracellular part of TF have function of intracellular signal conduction,which can promote the recording and composing of(VEGF).Research show VEGF participate the occurrence of UC,and which have relation with severity and scope involved.TF can promote expression of(MMP),MMP participate in tissue repair.It is demonstrate definitely MMP participate in inflammatory tissue repair,blood vessel formation and leucocyte chemotaxis in the patient with UC,which express highly in UC.TFPI can inhibit thrombus formation caused by TF that acted as the only physiological inhibitor of TF-FVIIa complex,prevent coagulation expanding.Clinical experiment show blood TFPI in patient with UC is higher than that in health,and that in active stage is higher than in ease.In this research,we detect the expression of TF and TFPI with each thirty cases in tissue of UC and normal colon using immunohistochemical method.The result is expression of TF is higher in tissue of UC,there is significant difference comparing with the expression in tissue of normal colon;TFPI is higher in tissue of UC,there is significant difference comparing with the expression in tissue of normal colon.Conclusion1.TF express commonly in cell membrane and cytoplasm in the mucosa near the ulcer,positive rate of TF expression in tissue of UC is remarkably higher than in tissue of normal colon.2.TFPI express commonly in cytoplasm in the mucosa near the ulcer with small brown granules in that,positive rate of TFPI expression in tissue of UC is remarkably higher than in tissue of normal colon.
