Objective:CD4~+CD25~+regulatory T cells(Treg)is an important CD4~+ regulatory T cells subgroup.This kind of cell subgroup possesses the characteristic of immunosupression and immunopotency. It's mechanism of action maybe through direct contact between cells and(or) secreting suppressor cell factors. It's important in maintaining immune homeostasis,preventing autoimmune disease, guiding transplantation tolerance. CD4~+CD25~+ T cells also have been reported to be potent inhibitors of an antitumor immune response.There are closely relation between the occurrence and growth of tumor and cellular immunity. Through detecting CD4~+CD25~+ T cells in peripheral blood of tumor patients,the current study was designed to investigate immune state of tumor patients,and the effects of CD4~+CD25~+ T cells levels after radiotherapy,chemotherapy,biotherapy, providing some beneficial data for tumorous combined therapy.Methods:The proportion of CD4~+CD25~+ T cells and CD3~+,CD4~+,CD8~+Tcells in the total amount T cells of 6 kinds of malignant tumor patients was evaluated by flow cytometry analysis. Peripheral blood of 194 malignant tumor patients were collected and the levels of CD4~+CD25~+ T cells were detected.60 healthy people for examination were selected at random as normal control.We observed disparation of the CD4~+CD25~+ T cells levels,and analysed the diversify and significance of the tumor patients' CD4~+CD25~+ T cells levels with different clinical stage. We analysed the influence and significance of patients' CD4~+CD25~+ T cells levels after radiotherapy ,chemotherapy and CIK biotherapy. Peripheral blood of tumor patients with empty stomach who were NSCLC,breast cancer, lymphoma and NPCwere collected 1 day before therapy,the 10th day after 2 periodicities chemotherapy and the 14th day after 2 course of CIK biotherapy.NPC patients were irradiated with 6MV X ray, peripheral blood of NPC patients were collected when the dose of radiotherapy was 50Gy,70Gy.49 breast carcinoma patients were administrated with 2 periodicities chemotherapy which were docetaxel and epirubicin.68 patients with lung cancer were administrated with 2 periodicities chemotherapy which were docetaxel and Cisplatin.Results:1. We found malignant tumor patients' levels of CD4~+CD25~+ T cells in peripheral blood were obviously higher than normal controls,P<0.05。2. The levels of CD4~+CD25~+ T cells in breast carcinoma patients were concerned with staging of tumor, with the higher of tumor staging,the levels of CD4~+CD25~+ T cells become higher,which was significant,P<0.05; The levels of CD4~+CD25~+ T cells in peripheral blood of gastric carcinoma patients , NSCLC patients with stageⅢ,Ⅳwas higher than those of patients with stageⅠ,Ⅱ,P< 0.05.3.The levels of CD4~+CD25~+ T cells in peripheral blood of NSCLC patients and breast carcinoma patients degrades after chemotherapy,the contrast was significant.There was no significant difference in levels of CD4~+CD25~+ T cells in peripheral blood before chemotherapy in the groups of sensitive to chemotherapy and no sensitive to it.The levels of CD4~+CD25~+ T cells of breast carcinoma had significant relation with metastatic lymph node,the number of metastatic lymph node.4.When the dose of radiotherapy was 50Gy,70Gy,the levels of CD4~+CD25~+ T cells in peripheral blood of NPC patients were significantly higher than the levels before radiotherapy,P<0.05,it was positive correlation with dose of radiotherapy,however, there was no obvious difference in the levels of CD3~+, CD4~+,CD8~+T cell and CD4~+/CD8~+in peripheral blood of tumor patients contrasting with the levels beforeradiotherapy.5.The levels of CD4~+CD25~+ T cells in peripheral blood of lymphoma patients degraded after CIK biotherapy, the difference was significant,P<0.05.The levels of CD4~+CD25~+ T cells in CIK cells were lower than the levels of it in peripheral blood before therapy, the difference was significant,P<0.05. Conclusion:1 .The levels of CD4~+CD25~+ T cells of NSCLC,breast cancer, lymphoma ,gastric cancer,ovarian cancer and NPC patients were significantly increased in peripheral bloodcompared with control group,and the cell immunity was seriously suppressed.2.The expression of CD4~+CD25~+ T cells in patients with NSCLC,breast carcinoma,gastric cancer was related with stage of TNM,the later of TNM,the higher expression ofCD4~+CD25~+T cells.3.The levels of CD4~+CD25~+ T cells in the peripheral blood of patients withNSCLC,breast carcinoma were decreased after chemotherapy,the difference wassigniflcant.It was benefit to strengthen immune function.There was no obviousdifference in levels of CD4~+CD25~+ T cells in peripheral blood before chemotherapy inthe groups of whether sensitive to chemotherapy or not.The levels of CD4~+CD25~+ Tcells can't be the factor of prediction.The levels of CD4~+CD25~+ T cells of breastcarcinoma had obvious relation with the number of metastatic lymph node .4.The levels of CD4~+CD25~+ T cells of NPC patients stepped up after radiotherapy,andwhich was positive correlated with dose of radiotheraphy.Radiotherapy may damagethe immune function of NPC patients.5.The levels of CD4~+CD25~+ T cells in peripheral blood of lymphoma patientsdegraded after CIK biotherapy,the contrast was significant.It enhanced the immunity of organism efficiently.We can conclude that one of the possible effective ways of CIK biotherapy is degrading the levels of CD4~+CD25~+ T cells in peripheral blood.
