Objective:To explore the effects and the possible mechanisms of plateled derived growth factor(PDGF) on neuroprotective after spinal cord injury. Hoping to provide a new idea and further a new method for the cure of spinal cord injury.Methods:90 SD rats were randomly divided into 3 groups:False traum group (group A,n=30),and nerval traum group(group B,groupC;each group n=30);GroupB(given saline in injured spinal cord),Group C(given PDGF in injured spinal cord).CBS were assessed by blinder observers at 1d befor surgery and 1d,7d,14d after injury.On the 3d,7d and 14d,3 experimental animals from each group were sacrificed for harvesting T7-T9 spinal tissue specimens.Taking the specimens for HE staining, observe the histological changes.Observe cell apoptosis and necrosis by transmission electro microscope.On the 1h,6h and 24h after injury,3 experimental animals from each group were sacrificed for harvesting T7-T9 spinal tissue specimens.Taking the specimens for immunohistochemistry and counting FLIN.Results:1,The evaluation of CBS scores showed:there was significant difference between group B and group C at each phase(P<0.05)2,Histological examination:In the group B a,3d after spinal cord injury,there were splinter haemorrhage in the gray mater and white mater and the damage of nerve cell;7d after spinal cord injury,there were proliferation of glia cell,endotheliocyte and lympho in the zone of injury; 14d after spinal cord injury,the proliferation is obvious.In the group C, the condtion of nervous tissue was better compared with group B;14d after spinal cord injury,the glia cell in the zone of injury are more than the group B.Neuro cell apoptosis and necrosis can be observed by transmission electron microscope.3,c-fos positive cells reach its peak at 1h after injury and decreased as time elongated,but it increased as time elongatd at 6h after injury and then reach its another peak at 24h after injury,positive cells in the groupC were apparently less than it's counterpart in the group B.Expression of c-fos between group C and group B were significant difference(P<0.05)Conclusions:1,By observing spinal cord morphology and the evaluation of CBS scores,we found that the PDGF has neuroprotecion effects in injured rat spinl cord.2,PDGF can attenuate the expression of c-fos,this means that PDGF can protect nerve cell and inhibit insecondary injury after SCI, this can be one of the mechanisms of PDGF.
