| Objective To investigate the effect of SOCS3 in the Pathogenesis of intrahepatic cholestasis of pregnancy(ICP).Methods The samples were obtained from 44 cases of ICP patients (ICP)including mild group(MG)(n=23)and severe group(SG)(n=21),25 cases of normal pregnancy women(NG).The expression and localization of SOCS3 in placentus were examined by immunohistochemical method.The level of IL-10 and IFN-γ,from placenta homogenation were measured by ELISA.Results(1)The immunoreactivity of SOCS3 was mainly found in the intracytoplasm of trophocyte.(2) The rate of positive cells and average gray scale of SOCS3 in placentus in NG,in MG and in SG were[(0.69±0.12),(81.06±6.79)],[(0.42±0.09),(147.29±7.11)],[(0.15±0.08),(203.85±6.58)],respectively. In NG,in MG and in SG the rate of positive cells were step down by tums,the average gray scale were step up by turns(P＜0.001).(3) The level of IL-10 and IFN-γfrom placenta homognation in NG,in MG and in SG were[(1.39±0.08)ug/L,(10.52±0.32) ug/L],[(1.22±0.75) ug/L,(13.41±0.47)ug/L],[(1.16±0.68)ug/L,(16.84±0.69)ug/L],respectively. The ratio of IFN-γ/IL-10 were(8.56±0.14),(11.65±0.19),(16.02±2.79), respectively.In NG,in MG and in SG the level of IL-10 were step down by turns,but the the level of IFN-γand the ratio of IFN-γ/IL-10 were step up by turns(P＜0.001).(4)There were negative correlation between SOCS3 and IFN-γ/IL-10 in ICP(P＜0.001).Conclusion1.The expression of SOCS3 in placentus in ICP group were statistically significantly lower than those in control group.SOCS3 may play a role in the pathogenesis of ICP and contribute to the development of ICP.2.There were negative correlation between SOCS3 and the ratio of IFN-γ/IL-10 in ICP.SOCS3 may play a role in the immunological regulation of Th1/Th2 in ICP.
