Objective:To observe the effects on growth inhibition and apoptosis of nasopharyngeal(NPC) cell lines CNE-1 and CNE-2,treated with sigular and combinative administration of TNF-related apoptosisinducing ligand(TRAIL) and Paclitaxel,to explore a brand new way for NPC combinative chemotherapy.Method:NPC cell lines CNE-1 and CNE-2 were treated by separate TRAIL and Paclitaxel at a variety of concentrations,growth inhibition rate at different time was tested by CCK-8 assay.2,NPC cell lines CNE-1 and CNE-2 were treated with seperate and combinative applications of TRAIL and Paclitaxel at the concentration of IC30 for 24h,and then growth inhibition rate was detected by CCK-8 assay,whole apoptosis rate and apoptosis rate in early and late stage were tested by flow cytometry.Result:1.In certain range of time and concentration,TRAIL and Paclitaxel inhibited the growth of CNE-1 and CNE-2 cells in a time and dose dependent manner.Their inhibition effects reached at a platform phase at a high drug concentration of TRAIL 1000ug/L and Paclitaxel 1×10-5mol/L,and the increasing tendence of growth inhibition rate was not apparent when the drug duration exceeds 48hs.2.The growth inhibition rate in TRAIL and Paclitaxel combinative group is more significant than the sum of which in the TRAIL and Paclitaxel sigular groups(P<0.05).3.The IC50 of TRAIL on CNE-1 and CNE-2 was about 100μg/L and 1000μg/L respectively,CNE-1 cell was sensitive to TRAIL while CNE-2 cell was opposite.4.For both CNE-1 and CNE-2 cells, there were statistical significance in all whole apoptosis rate and apoptosis rate in early and late stage in the combinative group,compared to the sum of apoptosis rate in both TRAIL and Paclitaxel sigular group(P <0.05).Conclusion:1.Sigular treatment of TRAIL and Paclitaxel can inhibit the growth of CNE-1 and CNE-2 cell line and induce apoptosis.TRAIL and Paclitaxel shows a synergistic effect while administrated together.2. TRAIL and Paclitaxel combinative therapy can enhance the drug sensitivity of CNE-2 cell to TRAIL,therefore sensitize CNE-2 to the killing effect of TRAIL,which provides a novel and prospective strategy for NPC combinative therapy.
