Objective In this study, the roles of IL-23 and IL-17 in EAN pathogenesis were explored and the molecular mechanism of the Rho kinase inhibitor was probed into by detecting the dynamic expression of IL-23p19 and IL-17 mRNA in the sciatic nerve, Peripheral blood mononuclear cells and lymph nodes during the course of experimental allergic neuritis rats and evaluating the changes of the expression under the influence of Rho-kinase.Methods 54 female Lewis rats were divided into 3 groups at random: EAN group (n=6, 6, 6) , EAN+Rho kinase inhibitor fasudil group(ROKi group) (n=6, 6,6) and CFA group (n=6, 6, 6) .The EAN rats were immunized with the component of Synthetic PNS myelin sheath protein P2 53-78aa and Freund's complete adjuvant. The ROKi group were injected intraperitoneally with Fasudil(25mg/kg.d) daily from 2-days long before the antigen immunization. The rats were executed at the 9th day, 17th day and 26th day from the day for immunization. The clinical symptoms of rats and the changes of demyelination and inflammatory cells infiltration of the sciatic nerves were evaluated. The expression of the IL-23p19 and IL-17 mRNA were detected by RT-PCR in different tissues, such as sciatic nerves, peripheral blood mononuclear cells and lymphonodes.Results The peak of clinical course arrived at the 17thd p.i in EAN rats, the expression of IL-23p19 and IL-17 mRNA were both higher on the9thd and the 17thd than the 26thd.p.i (p<0.05). There were significant differences between the EAN group and the CFA group at every time point (P<0.05);The demyelination and inflammation cells infiltration were ameliorated in spinal nerve. Compared with the EAN group, the clinical scores of the ROKi group were decreased markably (p<0.05); The expression of IL-23p19 and IL-17 mRNA in ROKi group were lower than EAN group at every time point (p<0.05).CFA group didn't show any clinical manifestation. The expression of IL-17 mRNA was positive corelation to that of IL-23p 19 (r=0.847, p<0.05) .Conclusions IL-23p19 and IL-17 might play a role in the pathogenesis of EAN, involved at the inducing and effect stage. In the EAN rats, the expression of IL-17 mRNA was corelation to that of IL-23p19mRNA. Rho-kinase inhibitor fasudil might ameliorate the development of EAN through down regulating the expression of the IL-23p19 and IL-17 mRNA.
