| Objective:To examine the expression of FADD and FLIP ,two apoptosis-related genes, in epithelial ovarian cancer,benign ovarian tumor and normal ovarian tissue, and investigate their chinical significance.Methods: tissue specimens from 44 cases of primary ovarian cancer,25 cases of benign ovarian tumor and 25 cases of normal ovarian tissue were examined by antibody of FLIP and FADD with immunohistochemical technique(SP staining, streptomycin-avidin-peroxidase) were used to examine the expressine among FLIPand FADD in tumor tissues . Their differences of FLIP and FADD expression among the three groups were assessed by Pearson Chi-Square test. Immunohistochemical method(SP staining, streptomycin-avidin -peroxidase) and Reverse transcription-polymarase chain reation(RT-PCR)were used to examine the expression of FLIP and FADD in tumor tissues. The correlations between FLIP and FADD expression and several clinicopathological parameters,such as the type of pathology, the grade of histopathology and clinical stage,were assessed by Pearson Chi-Square test, too.Results:1. the expression rate of FLIP in different ovarian tissue was : in the primary epithelial ovarian cancer, the benign ovarian tumor and the normal ovary tissue ,the expression rate of FLIP was 68.18%, 28.00% and 32.00%, the difference was significant via the Pearson chi-square test (P<0.001). The expression rate of FLIP was 30.00% in Clinical stage I-II, the expression rate was 79.41% in Clinical stage III-IV, the difference was significant via the Fisher's exact probabilities test (P =0.006, P<0.05). The expression rate of FLIP was 47.62% in histology grade 1, the expression rate was 86.96% in histology grade 2-3, the difference was significant via the Pearson chi-square test (P =0.005). in the serous cystadenocarcinoma, the expression rate of FLIP was 54.55%, in the mutinous adenocarcinoma,the expression rate was 59.09%, in the others, the expression rate was 66.67%, the difference was not significant via the Pearson chi-square test (P=0.960).2. in the primary epithelial ovarian cancer, the benign ovarian tumor and the normal ovary tissue ,the expression rate of FADD was 59.09%, 84.00% and 80.00%, the difference was significant via the Pearson chi-square test (P=0.027). The expression rate of FADD was 100.00% in Clinical stage I-II, the expression rate was 67.65% in Clinical stage III-IV, the difference was significant via the Fisher's exact probabilities test (P =0.046). The expression rate of FADD was 71.43% in histology grade 1, the expression rate was 39.13% in histology grade 2-3, the difference was significant via the Pearson chi-square test (P =0.032). In the serous cystadenocarcinoma, the expression rate of FADD was 59.09%, in the mucinous adenocarcinoma,the expression rate was 57.89%, in the others, the expression rate was 66.67%, the difference was not significant via the Pearson chi-square test (P=0.960).3. The value of FLIP protein expression was 1.02±0.17 in histology grade 1, the value was 1.33±0.18 in histology grade 2-3, The value of FLIP mRNA expression was 2.07±0.19 in histology grade 1, the value was 2.34±0.34 in histology grade 2-3; The value of FADD protein expression was 1.56±0.39 in histology grade 1, the value was 1.19±0.20 in histology grade 2-3, The value of FADD mRNA expression was 3.65±0.38 in histology grade 1, the value was 2.85±0.41 in histology grade 2-3; The value of PCNA protein expression was 1.43±0.21 in histology grade 1, the value was 1.71±0.20 in histology grade 2-3.the difference was significant (P<0.05).The value of FLIP protein expression was 1.00±0.24 in clinical stage I-II, the value was 1.26±0.21 in clinical stage III-IV, The value of FLIP mRNA expression was 1.89±0.01 in clinical stage I-II, the value was 2.27±0.30 in clinical stage III-IV; The value of FADD protein expression was 1.76±0.40 in clinical stage I-II, the value was 1.21±0.17 in clinical stage III-IV, The value of FADD mRNA expression was 3.49±0.25 in clinical stage I-II, the value was 3.01±0.42 in clinical stage III-IV; The value of PCNA protein expression was 1.28±0.13 in clinical stage I-II, the value was 1.65±0.21 in clinical stage III-IV.the difference was significant (P<0.05). Conclusions:1. The positive expression rate of FLIP was high in epithelial ovarian cancer.the higher histopathology grade,the later clinical stage, the higher the positive expression rate of FLIP. It illustrate that the high expression rate of FLIP may play an important role in the accurrence and development of epithelial ovarian cancer.2. The positive expression rate of FADD was low in epithelial ovarian cancer.the higher histopathology grade,the later clinical stage, the lower the positive expression rate of FADD. It illustrate that the low expression rate of FADD may play an important role in the accurrence and development of epithelial ovarian cancer. |
