Objective: To observe the effects of intermittent hypoxia training and high-fat diet to the myocardial morphology of mice, and investigate whether intermittent hypoxia training have protection to the myocardial morphology damages caused by high-fat diet. Methods: 24 female KM rats were divided into 4 groups randomly: the normal diet group (group CON), the intermittent hypoxia training group (group IHT, 3000m altitude, 0.07MPa, FiO2 14%, 15min hypoxia/5min normoxia, 9 cycles/d), the high-fat-diet group (group HFD, 10% lard, 10% eggs, 5% sugar, 75% basement forage), the high-fat-diet company with intermittent hypoxia training meanwhile (group HFD+IHT). During the 40 days, observed the weight changes of each group, After 40 days, compared the food and energy consumes of each group, then the myocardium of left ventricular was harvested, observed the changes of morphology with HE staining under light microscope, evaluate the ultrastructure changes under TYPE H7500 electron microscope. And the changes of mitochondria and myofibril were also assessed by score 0-4 grade semiquantitatively per sample and received Flameng semi-quantitative analysis. Results: Compare the group CON, the other groups showed morphology changes, but the group IHT and HFD+IHT had slighter morphology changes compare the group HFD. And in score of mitochondria and myofibril were showed the same results. Mitochondria scores (CON 0.810±0.689, IHT 1.740±0. 770, HFD+IHT 1.800±0.664, HFD 2.240±0.650). Myofibril scores (CON 0.800±0.761, IHT 1.430±0.679, HFD+IHT 1.700±0.750, HFD 3.370±0.490). IHT vs HFD+IHT p>0.05, IHT&HFD+IHT vs HFD p<0.001.Conclusion: It was suggested that the high-food-diet have damages to the morphology of myocardium, and these damages decreased when intervened by intermittent hypoxia training.
