ObjectiveTo investigate the expression of Nogo-A and GAP-43 proteins following intracerebral hemorrhage and intervention with Edaravone after ICH in rats and explore the neuroprotective effect of edaravone on experimental intracerebral hemorrhagic rats.MethodsOne hundred and twenty Sprague Dawley(SD) rats were randomly divided into three groups:the sham-operated group(group A,n=40),the intracerebral hemorrhage(ICH) group(group B,n= 40),the intracerebral hemorrhage plus edaravone(6mg/kg) intervention group(group C,n = 40).Every group was divided into 5 subgroups:3rdd,7thd,14th d,21std and 28thd after operation;Animals were sacrificed at the corresponding time points;the subgroups of 3rdd,7thd,14thd contain 10 rats,half of which were used to measure brain water content and the other for neuroethology assessment and immunohistochemical detection;subgroups of 21std and 28thd include 5 rats,which were used for neuroethology assessment and immunohistochemical detection.In group B and group C the ICH models were established by a microinfusion pump,containing 2μl saline and 0.5u collagenaseⅦ,stereotaxically injected into right caudate nucleus;group A only received 2μl saline.6 hours after ICH the rats in group C were intraperitoneally injected edaravone 6mg/kg,qd,the same treatment continued from the 1st to the day when the rats were sacrificed;group A and group B only received equal amount of saline.Longa tests and Forelimb Placing tests were performed to value neurological function and brain water content was determined by dry-wet weight method,the expression of Nogo-A and GAP-43 proteins in brain tissu were examined by immunohistochemical ABC method and image analysis.ResultsCompared with group A,in group B,the experssion of Nogo-A gradually increased from 3rdd to 28thd,and higher than that of sham-operated group(P<0.05,P<0.05,P<0.05,P<0.05,P<0.05);GAP-43 proteins were strongly experssed at 3thd and reached the peak at 7thd, gradually decreased from 7thd to 28thd,expression of GAP-43 was higher than that of group A at 3rdd,7thd,14thd and 21std(p<0.05,P<0.05,P<0.05, P<0.05),there was no statistical difference at 28thd(P>0.05).In group C the experssion of Nogo-A gradually increased from 3rdd to 28thd,which higher than that of group A(P<0.05,P<0.05,P<0.05,P<0.05,P<0.05);GAP-43 was strongly experssed at 3thd and reached the peak at 7thd, gradually decreased from 7thd to 28thd,expression of GAP-43 was higher than that of group A at every time point(P<0.05,P<0.05,P<0.05,P<0.05, P<0.05).Compared with group B,in group C neurological deficit score has significantly improved at 3rdd,7thd,14thd,21st d and 28th d(P<0.05,P<0.05,P<0.05,P<0.05,P<0.05);water content in brain tissue were significantly lower than that in group B at 3rdd and 7thd(P<0.01,P<0.01),there was no significant difference at 14thd(P>0.05);the experssion of Nogo-A showed no significant difference at 3rdd,7thd and 14thd(P>0.05, P>0.05,P>0.05)and a significant reduction at 21std and 28thd(P<0.05,P<0.05);the experssion of GAP-43 showed no significant difference at 3rdd and 7thd(P>0.05,P>0.05) and a significant increase at 14thd,21std and 28thd(P<0.05,P<0.05).Conclusions1.Edaravone can effectively alleviate brain edema and accelerate the recover of neurological function deficit after ICH.2.Edaravone probably decrease the expression of Nogo-A,prolong the expression of GAP-43 and ultimately acted neuroprotective effects...
