Objective: To find a modified method for primary culture of human eccrine sweat gland epithelial cells, which cost less operating procedure and time than traditional culture methods, so as to obtain more viable sweat gland epithelial cells in shorter time. And thus to provide theoretical and practical basis for the research of sweat gland and functional tissue engineered skin.Methods: Based on the traditional tissue block culture method of sweat gland, we make the sweat gland pieces stick onto the bottom of the culture dish stably by place a cover slip upon them, keep the pieces from floating up when changing medium or doing other operations. By doing so, experimental operation could be simplified because media exchange frequency was reduced.Results: The modified method enhanced the adherent rate of the eccrine sweat gland tissue pieces significantly, meanwhile had no impairment to theirs proliferative ability, so that could obtained more sweat gland epithelial cells than the traditional methods in the same amount of time.Conclusion: Compared with the traditional method, this method simplified the laboratory procedure and shortened the experimental time consume, cultured cells own the superiority of quantity. The cultured human eccrine sweat gland epithelial cells had still maintained the epithelial cells'structural characteristics, moreover, they had well viability, all of these indicate the feasibility of construct eccrine sweat gland by tissue engineering.
