Effect Of Perindopril On The Expression Of Cx43 And MMP-7 In Infarct Border Zone

Objective To observe the expression of connexin43(Cx43) and Matrix Metalloproteinase-7(MMP-7) in infarct border zone, and investigate the effect of perindopril on the expression of Cx43 and MMP-7 in infarct border zone, to explore the effect of perindopril on the expression of Cx43.Methods 30 adult male Sprague-Dawley rats were randomly separated into three groups: sham operation group (S group), operation group(O group) and operation+ perindopril group(P group). myocardial infaraction(MI) rat model was established in the experiments by ligation of left anterior descending(LAD) coronary. one day after operation, all rats were raised for 2 weeks and perindopril 2mg/(kg·d) were added to rats in P group. The expression of Cx43 and MMP-7 in heart tissue was determined by immunofluorescence(IF).Results Compared with S group, the Cx43 protein level of ventricular tissue in O group decreased significantly with misdistribution and the MMP-7 protein level of ventricular tissue in O group increased significantly(P<0.05). Compared with O group, the Cx43 protein level in P group increased significantly, the MMP-7 protein level in P group decreased significantly(P<0.05) and the inhomogeneity in Cx43 distribution decreased.Cx43 and MMP-7 in O group and P group were negative correlation in infarct border zone. (P<0.05, r=0.932)Conclusion In infarct border zone, the expression of Cx43 protein decreased significantly with misdistribution and the expression of MMP-7 protein increased significantly. They seemed to be negative correlation. Perindopril increases the expression of Cx43 protein, attenuates Cx43 remodeling effectively and inhibits the excessive expression of MMP-7 protein. Increasing of Cx43 protein expression maybe correlated with inhibition the excessive expression of MMP-7 protein.
