| Objective: To analyze the clinical data and auxiliary examination of tuberculosis in Chongqing. To investigation the correlation between the polymorphisms of IL-27 (-964A/G and 2905T/G), IL-18Rα(-69C/T and -638C/T) gene and susceptibility to tuberculosis and clinical manifestation in Chongqing Han Population. To reveal the influence of those polymorphisms to tuberculosis susceptibility.Methods: To overview the clinical data and auxiliary examination of the 385 TB patients in Chongqing Han Population. The polymorphisms of IL-27 (-964A/G and 2905T/G) gene in 385 patients with tuberculosis and 391 healthy controls and IL-18Rα(-69C/T and -638C/T) gene in 427 patients with tuberculosis and 469 healthy controls were detected by using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and sequence specific primers polymerase chain reaction (PCR-SSP), and their susceptibility to tuberculosis were analyzed.Results:1. Statistically significant differences in types were found between children patients and adults patients with tuberculosis. The primary pulmonary tuberculosis and extrapulmonary tuberculosis were predominent in children, while in adults the secondary tuberculosis was predominant. The ratio of cerebral tuberculosis in children was higher than in adults (χ~2=61.446, P=0.001). The ratio of cerebral tuberculosis who did not vaccinate BCG is higher than who did (χ~2=4.264, P=0.039). The ratio of cerebral tuberculosis who exposed to tuberculosis was higher than who did not (χ~2=4.538, P=0.033).2. The polymorphisms of -964A/G in IL-27 gene: The frequencies of AA genotype and A allele at IL-27 -964 site were significantly higher in each TB groups than their healthy controls of children group, adults group and total group, respectively (P<0.05). The genotype frequency of AA of -964A/G in those with positive yield was significantly higher than those with negative yield (χ~2=7.152, P=0.03). There were no differences in AG and GG genotypes frequencies of IL-27 -964A/G between patients with tuberculosis and controls in children group, adults group and total group, respectively.3. The polymorphisms of 2905T/G in IL-27 gene: There were no differences in alleles and genotypes frequencies of IL-27 (2905T/G) between patients with tuberculosis and controls in children group, adults group and total group, respectively (P>0.05).4. The polymorphisms of -69C/T in IL-18Rαgene: The frequency of IL-18Rα-69CC genotype in tuberculosis group was significantly lower than that in controls of children group, adults group and total group, respectively (P<0.05). The frequencies of -69TT genotypes were significant higher in tuberculosis than in controls of children group and total group, respectively. Allele T at -69 was of a significantly higher frequency in tuberculosis than that in normal controls (P<0.05). And there was significant higher of alleles T at -69 in cerebral tuberculosis than other tuberculosis out of cerebral tuberculosis (χ~2=4.064, P=0.044).5. The polymorphisms of -638C/T in IL-18Rαgene: The frequencies of all genotypes or alleles at IL-18Rα-638 site were not significant difference in tuberculosis and normal controls in children group, adults group and total group, respectively (P>0.05).Conclusion:1. Statistically significant differences in the tuberculosis types were found between children patients and adults patients. Children are more susceptible to cerebral tuberculosis. To prevent tuberculosis especially cerebral tuberculosis one of the most important questions is BCG vaccination suscessfully and refraining from exposure to tuberculosis.2. This study confirmed the association between the polymorphisms -964A/G of IL-27gene and susceptibility to tuberculosis,and with A allele is more susceptibility to tuberculosis and positive yield in Chongqing Han Population. Allele G may plays a role in preventing tuberculosis infection. There is no association between IL-27 (2905T/G) polymorphisms and susceptibility to tuberculosis in Chongqing Han Population.3. There have the association between the polymorphisms of -69C/T in IL-18Rαgene and susceptibility to tuberculosis in Chongqing Han Population,and tuberculosis patients with T allele is more susceptibility to cerebral tuberculosis. Alleles C of position -69 possibly play a role in preventing tuberculosis. But -638C/T polymorphisms of IL-18Rαgene is not related with tuberclosis of Chongqing Han Population possibly. |
