Objective:To assay fasting plasma preptin level by radioimmunoass- ay in the patients with different glucose tolerances. And investigate the relationship between plasma preptin levels and body mass index(BMI), waist hip ratio(WHR), blood glucose, plasma insulin levels, lipids as well as other factors.Methods:160 subjects were divided into three groups. 50 patients with type 2 diabetes mellitus(T2DM), 56 patients with impaired glucose tolerance(IGT), they were all preliminary diagnosis. 54 normal control subjects(NGT) through health examination in our clinic service.Collected fasting and two hours venous blood of these subjects which be given 75 gram amylaceum oral application, then measured the levels of plasma glucose, plasma insulin, glycosylated hemoglobin, lipids, preptin by different methods. The relationship between fasting plasma preptin levels and body mass index, waist hip ratio, plasma glucose, plasma insulin, lipids, insulin resistance index were analyzed by statistical package for the socialsciences.Results:Fasting plasma preptin levels in T2DM were significantly increased compared with IGT and NGT[(456±14)ng/L vs(416±13)ng/L and(398±13)ng/L, p<0.05]. Fasting plasma preptin levels correlated positively with DBP, TG, TC, HDL-C, FFA, 2hBG, HbA1C and HOMA-IR [r=0.20, r=0.24, r=0.24, r=0.18, r=0.21, r=0.18, r=0.19, r=0.13, P<0.01, P<0.01, P<0.01, P<0.05, P<0.01, P<0.05, P<0.01, P<0.05] in simple reg- ression analysis. DBP, TG, HDL-C and FFA were independently related with fasting plasma preptin levels, the equation was:Y=147.959 +1.876XDBP+17.827 XTG+47.869X HDL-L+59.964X FFA. Multiple logistic regress analysis revealed preptin was correlated positively with type 2 diabetes.Conclusion:The changes of fasting plasma preptin levels maybe associate with glucose and lipid metabolism disorder, and it may contribute, in part, to the pathogenesis of insulin resistance and type 2 diabetes.
