AIM : To investigate the effect of hydroxychloroquine(HCQ) on ventricular remodeling after myocardial infarction ( MI ) in rats. METHODS:MI and sham-operation were established by Maisel's method. Three days after operation, surviving Sprague-Dawley(SD)rats were divided into MI hydroxychloroquine group(MI+HCQ,n=11)and MI placebo group(MI,n=15), sham-operation hydroxychloroquine group(SHAM+HCQ,n=7) and sham-operation placebo group(SHAM,n=7), which were treated with hydroxychloroquine and physiological saline respectively. Mortality rate of each group was calculated from three days to eighty seven days after the operation. Cardiac structure index(IVSd,LVPWd,LVDd)and function index(FS,EF) were examed by Doppler ultrasonograph.Protein expression of P-Akt and P53 around the infarction zones was detected by immunohistochemistry and Western blot. Apoptosis was observed by TdT-Mediated dUTP nick end labeling (TUNEL). RESULTS:Mortality rate in MI+HCQ group, MI group, SHAM+HCQ group , SHAM group were 9% ,40% , 0% , 0% respectively. In MI group,IVSd,LVPWd grew thicker and then became thinner, LVDd grew bigger, FS,EF became smaller;in MI+HCQ group,IVSd,LVPWd grew thicker, LVDd remained the same, FS,EF grew bigger. Eighty seven days after operation,in MI+HCQ group and MI group, IVSd were (0.124±0.009) cm,(0.117±0.007) cm (P <0.05);LVPWd were (0.15±0.009) cm, (0.125±0.008) cm(P <0.05);LVDd were (0.636±0.012) cm ,(0.72±0.013) cm (P <0.05);FS were (29.44±2.13) % ,(20.39±1.83) %(P <0.05);EF were (62.23±2.33) % ,(47.99±1.89) %(P <0.05);IOD of P-Akt were(1.12±0.21),(0.58±0.18)(P <0.05);IOD of P53 were (1.01±0.20), (0.98±0.19)(P >0.05). Apoptosis rate were12±4%,30±5%.In SHAM+HCQ group and SHAM group ,IVSd were (0.128±0.010) cm, (0.122±0.008) cm ( P <0.05) ; LVPWd were ( 0.152±0.010) cm, (0.137±0.009) cm(P <0.05);LVDd were (0.596±0.011) cm, (0.574±0.012) cm (P >0.05);FS were (34.33±2.13) %, (35.57±1.78) %(P >0.05);EF were (69.82±2.91) % ,(71.15±2.11) %(P >0.05);IOD of P-Akt were (0.78±0.20), (0.62±0.23)(P >0.05);IOD of P53 were (0.49±0.22), (0.32±0.19)(P >0.05). Apoptosis rate were2±2%,3±2%.CONCLUSION:Hydroxychloroquine can prevent ventricle remodeling after MI by modulating.
