Objective To investigate the expression of NF-κB and ICAM-1 in the myocardial of diabetic rat and the intervention of the rosiglitazone (RGZ).Methods The rats were divided into normal control (NC) group,model(M) group,RGZ group.The normal control group and the model group were orally administered the same dosage of saline. The RGZ group were orally administered rosiglitazone.After 8 weeks of drug intervention, the levels of FBG, TC, TG, HDL-C, LDL-C and FFA in serum. The protein contents of ICAM-1 were measured by IH. The protein contents of NF-κB were measured by IH and western blotting.Results (1) The levels of FBG, TG and FFA in the M group were significantly different from that of the NC group(P<0.01~P<0.05); Compared with the M group, the levels of FBG decreased(P<0.05),the levels of TC, TG, LDL-C, HDL-C and FFA were not significantly different from that of the RGZ group.(2) HE staining: Cadiocytes were fine in the NC group, and the muscle fiber was concinnous; In the M group, Cadiocytes were hypertrophy, and the muscle fiber was indiscriminate alike waves and ruptured; Compared with the M group, Cadiocytes were smaller and the muscle fiber was more concinnous in the RGZ group.(3) Compared with the NC group, the protein contents of NF-κB and ICAM-1 increased in the M group(P<0.01). The protein contents of NF-κB and ICAM-1 were significantly lower than those in the M group(P<0.05).Conclusions Rosiglitazone had decreased fasting blood glucose, improved indiscriminate muscle fiber in cardiac muscle, also decrease the protein contents of NF-κB and ICAM-1, but no obviously effect on blood lipids and free fatty acid. Thus rosiglitazone was effective on myocardial protection in diabetic rat by decreasing blood glucose, improving heart muscle fibrosis, ameliorating oxidative stress as well as inflammation activity in myocardial tissue.
