| Allium is one of the Liliaceae which commonly known as Congsuan category, and usually have a special smell. We have done many studies and have developed a variety of health food and medicines for anti-bacterial, anti-cancer and prevention, such as cardiovascular and cerebrovascular diseases. There is still great potential on the development of Allium plants. Fistular onion stalk play an important role in the pharmacological.Boxintong is a traditional Chinese medicine. It plays a role in anti-oxidative damage and can improve endothelial function. It can be used for prevention and treatment of atherosclerosis. Fresh fistular onion stalk is dried by microwave vacuum dry first, and then be processed by supercritical extraction. We can get yellow cream with a special smell. The cream is the raw materials for Boxintong.This study is to understand the chemical constituents of the cream. By modern separation and analysis technology, the cream has been studied. Nine compounds were isolated by using silica column chromatography and Sephadex LH-20 column chromatography, six compounds from which have been identified with the help of UV, IR, MS, 1H-NMR, 13C-NMR. The chemical structures of six compounds are: hexadecanoic acid(â… ), Paeonol(â…¡), neocerotic acid , tricosyl ester (â…¢) , 4-heptadecyl-3-henicosenol(â…£),β-tocopherol (â…§), di(ethylhexyl)phthalate (â…¨).We also isolate the cream into seven components by using silica column chromatography. And by GC-MS combining with computer database analysis, we find the differences between various components. We also compare the cream from fistular onion stalk and the cream that supercritical extracted from the leaves to understand the difference of the chemical composition between them.Reference best conditions of garlic biological enzyme, we set two groups, one has been enzyme hydrolyzed by adjusting temperature and PH , but kill the enzymes with methanol of the other group. With GC-MS analysis, we compare the type and content of organic sulfur compounds of the two groups. We found that the content of organic sulphur compounds was significantly improved in the experimental group. By area normalization method, we calculated the total percent of sulfur compounds are between 24%~28%. But only three sulfur compounds are detected from the control group, the total content is 2.98%. The major categories are thiophene, thiazole, disulfides, trisulfides and some ring of sulphur compounds. Many of these sulfur compounds are detected in garlic , they are important substances of Allium plants.By area normalization method, the total percent of sulfur compounds of the cream is more or less than 10%. If we enzymed fistular onion stalk before the supercritical processing, sulfur compounds can be achieved to 20% of total content .That solutions can greatly improve the content of organic sulphur compounds.Through this study we get a systematic understanding of chemical constituents of the raw material of Boxintong. And do some researches of enzymolysis. The best conditions of enzyme hydrolysis, the biological extraction process of Allinase, the mechanism of enzymolysis need for deeper study. |
