Study On The Lipid-lowering Therapy And Mechanism Of Compound Zhenzhu Tiaozhi Capsule On Hyperlipidemia Rats

Objective:To optimize the extracting process of compound zhenzhu tiaozhi capsule. 0n this basis,to observe the effect of lipid-lowering therapy and to probe the mechanism of compound zhenzhu tiaozhi capsule,in order to provide the scientific basis to clinical medicine.Methods:Rats model with hyperlipidemia were established with high fat emulsion by lavage. Meanwhile,drugs of the three different extracting process of compound zhenzhu tiaozhi capsule and fenofibrate were fed to every group respectively. After 4 weeks, serum cholesterol (TC), triglyeride (TG) were detected,and to select the optimal extracting process of compound zhenzhu tiaozhi capsule according to TC and TG levels. 0n this basis,rats model with hyperlipidemia were established with high fat emulsion by lavage for 2 weeks. Then,the serum TC were detected. According to serum TC,the hyperlipidemia rats were divided into model group,zhenzhu compound tiaozhi capsule high dose group,moderate dose group,low dose group,and the control groups(Xuezhikang capsule group, Fenofibrate group) at random. Rats except in normal group were fed with high fat emulsion in a.m.,and rats in medication groups were fed with corresponding drugs while the remaining rats were fed by equal saline in p.m..6 weeks later,to observe the lipids-lowering therapy and to probe the effects on apolipoprotein metabolism,lipid peroxidation,lipid metabolizing enzymes,vascular endothelial function,hemorheology,liver lipids and liver histopathology of compound zhenzhu tiaozhi capsule.Result:Extracting process A which can also reduce the serum TC and TG were the optimal extracting process for compound zhenzhu tiaozhi capsule.The serum TC of model rats fed high fat emulsion for 2 weeks was increased. It suggested that the hyperlipidemia model were established successfully.In model group which continued to be fed high fat emulsion for 6 weeks, the levels of serum TC,TG, low density lipoprotein cholesterol (LDL-C) were rised,and high density lipoprotein cholesterol (HDL-C) reduced.The results also showed that the levels of ApoAâ… ,SOD,LPL,HL were decreased,that the ApoB,MDA were increased.The NOS and NO were not significantly changed.The whole blood viscosity,plasma viscosity, casson viscosity were increased to induce abnormal hemorheology.Liver index,liver TC were elevated,and there were some pathological changes in liver such as liver tissue steatosis, hydropic degeneration,necrosis, inflammatory cell infiltration.6 weeks after administration,in compound zhenzhu tiaozhi capsule groups the levels of serum TC,TG,LDL-C,AI of were significangtly lowwed, and the HDL-C,HDL-C/TC were rised .The content of ApoAâ… ,ApoAâ… /ApoB, NO were increased and the activities of SOD,LPL,HL were enhanced. The whole blood viscosity,plasma viscosity, blood reduced viscosity were decreased to ameliorate hemorheology.The liver index and liver TC were reduced. Liver tissue steatosis, hydropic degeneration, necrosis, inflammatory cell infiltration were lightened,and pathological changes were significantly ameliorated.Conclusion: The optimum extracting process was extracting process A of compound zhenzhu tiaozhi capsule which showed certain therapeutic effect in hyperlipidemia rats induced by high fat emulsion.The lipid-lowering therapy may be due to its effects on regulating the metabolism of ApoAâ… and ApoB,enhancing the activity of important enzymes in lipid metabolism (LPL,HL) to regulate the metabolism of lipids, increasing SOD and reducing MDA to improve the ability to resist oxidation, raising the level of NO to improve endothelial function,ameliorating hemorrheology, reducing the deposition of liver lipids and ameliorating pathological impairment.
