Objective: To discuss the distribution characteristics of HLA-A,B and DRB1 genes and the polymorphism of fifteen short tandem repeat (STR) in human body at the critical life stage.Methods: We determined sixty-nine healthy and unrelated old people aged between 70 to 89-year old (including 37 males and 32 females) as the sample group, and two hundreds volunteer stem cell donators aged between 18 to 45-year old was the control group for HLA. Other one hundred and fifty non-related healthy adults (18 to 50 years old) were classified as the control group for STR. DNA was extracted from intravenous blood samples which were added with EDTA-K2. The typing test for HLA-A,B and DRB1 was performed by means of SSO-PCR. The typing test for 15 STR loci (CSF1PO, D7S820, D8S1179, D21S11, D2S1338, D3S1358, D13S317, D16S539, TH01, D18S51, D19S433, TPOX, VWA, D5S818 and FGA) was performed by means of capillary electrophoresis. The frequency distribution of alleles and the homozygote frequencies between the two groups were compared and analyzed.Results: Total 46 alleles at the loci HLA-A,B and DRB1 had been detected in the sample group, whose frequencies did not have significant difference comparing with that in control group (P>0.05). Total 131 alleles were detected in the typing test for 15 STR loci. The frequencies of alleles D18S51*21, D5S818*14 and FGA*22.2 in the sample group were higher than those in control group (P<0.05). The frequency of allele CSF1P0*12 was lower than that in control group (P<0.05). It was shown that by analysis of homozygote frequency of loci HLA-A,B and DRB1 and 15 STR loci, the actual homozygote frequency of loci CSF1PO and D13S317 in the sample group was higher than the theoretical frequency value.Conclusion: There may be anti-aging genes near the STR loci D18S51, D5S818 and FGA and there may be accelerated aging genes near to the locus CSF1P0. The fact that the actual homozygote frequency value of loci CSF1PO and D13S317 is higher than the theoretical frequency value suggests that the homozygote trend of such two STR loci may contribute to the survival of people at the life critical stage.
