Objective: To observe the effects of the method of invigorating kidney and regulating menstruation (MIKRM) on glycometabolism of PCO rats, to probe the mechanism of MIKRM in the treatment of glycometabolism disorder of Polycystic Ovarian Syndrome(PCOS).Methods: All rats were randomly divided into five groups: normal control group (NCG), model control group (MCG), invigorating kidney formulaâ… control group (IKFâ… CG), invigorating kidney formulaâ…¡control group(IKFâ…¡CG), invigorating kidney and regulating menstruation control group(IKRMCG). According to the physiological and pathological characteristics of PCOS combined with experiment requirements, PCO models were induced by progesterone and HCG. Each rat of NCG and MCG was treated with intragastric administration of distilled water of 20ml/kg?d; IKFâ… CG with invigorating kidney formulaâ… of 47.75g/kg?d; IKFâ…¡CG with invigorating kidney formulaâ…¡of 46.00g/kg?d; according to a 4-day estrous cycle of rat, IKRMCG with invigorating kidney formulaâ… , 47.75g/kg?d during the first two days; invigorating kidney formulaâ…¡, 46g.00g/kg?d in the next three and four days. After 16 days'treatment, body weight was weighed, the serum insulin levels were measured by RIA, the serum resistin levels were detected by ELISA for every rat, and the histo-pathology of ovarians were observed.Results: The pharmacological research has demonstrated that compared with MCG, body weight of IKFâ… CG and IKFâ…¡CG have obviously reduced and the difference had extremely remarkable statistical significance (P<0.01), the serum insulin levels have obviously reduced and the difference had the remarkable statistical significance (P<0.05), while serum resistance levels have reduced in varying degrees, but the difference has no remarkable statistical significance (P>0.05); body weight, the serum insulin and resistin levels of IKRMCG have obviously reduced and the difference had extremely remarkable statistical significance (P<0.01).Compared with IKRMCG, the serum resistin levels of IKFâ… CG and IKFâ…¡CG have obviously reduced and the difference had extremely remarkable statistical significance (P<0.01). It is manifested that MIKRM was able to obviously reduce the body weight, the serum resistin and insulin levels of model rats, indicating that it can improve insulin resistance and hyperinsulinemia, adjust the resistance level, improve obesity and regulate glycometabolism in PCO rats. It indicated that MIKRM is more effective than a separate application of invigorating kidney formulaâ… orâ…¡. MIKRM can thicken the layers of granular cell, make layers of follicle cell thinner, reduce the expansion of ovarian cyst and increase the numbers of corepus luteum. The repairation of ovarian morphology of IKFâ… CG and IKFâ…¡CG was not as good as IKRMCG. It showed that MIKRM was better at promoting ovulation and luteinizing development and improving polycystic ovary syndrome and ovarian functions of model rats.Conclusion: MIKRM can obviously reduce body weight, the serum resistin and insulin levels of model rats and can reduce the expansion of ovarian cyst, increase the layers of granular cells, which indicated that MIKRM can adjust glycometabolism disorders and improve the reproductive endocrine function of the ovary in PCOS. And the effect of MIKRM is far superior to a singular application of invigorating kidney formula.
