Objective:In this study,model of mesangial proliferativeglomerulonephritis(MsPGN) was made with immunization.The MsPGN rats were cured by the flavons in Decumbent Bugle Herb(FDBH).Proteinuria plasma protein,blood-fat,renal function,renal pathomorphology were observed.The effect and mechanism of DBH on MsPGN were investigated so as to provid experimental data.Methods:Sixty-eight male Sprague-Dawley rats were adopted and model of MsPGN was made with immunization except 10 rats which were randomly choosed as normal group.At the end of 5th week,the amount of 24h urinary protein was assayed to reject the minus rats.The rest rats were randomly divided into five groups:model group,Tripterygium glycosides (TPG)-treated group and(high,medium and low dose) FDBH-treated group.From 6 to 12 week,Treated group were feeded with TPG or FDBH(respectively,FDBH:2.16,1.08,0.54g·kg-1·day-1 ),TPG(0.018g·kg-1·day-1).In the end of experiment,the 24h urinary protein contents,the level of CRE,BUN,ALT,TP,TG,TC,MDA,SOD,IL-1,TNF-a and TGF-βwere determined,The pathological changes,ultrastructural organization and the immune complex deposits was observed.Result:1.Urinary protein:The amount of urinary protein was significantly increased in modle rats except 4 negative ones he end of 5th week(P<0.01,compared with normal group). After six weeks treatment,the urinary protein was decreased in the entire treatement group (P<0.01or 0.05,compared with model group).2.Biochemical item:After six weeks treatement,compare with normal group:the TP,TG, Tch,BUN,Cr,SOD,MDA,TNF-a,IL-1 was increased in modle group(P<0.05 or 0.01),but there was no significant difference in ALT compared with the control group(P>0.05).Compare with normal group:the TP,SOD was increased,and the MDA was decreased(P<0.05 or 0.01).The TG,Tch,BUN,Cr,TNF-a,IL-1 was decreased obviously in the TGP-treated group,high dose FDBH-treated group and medium dose FDBH-treated group. Compared with modle group,the ALT in each FDBH-treated groups has no signifcant difference(P>0.05),but was increased obviously in TGP-treated group(P<0.01).Compared with TGP-treated group:the effedt of decreasing TG and Tch was significant in the high dose FDBH-treated group(P<0.05 or 0.01).And the effect of decreasing Tch,MDA, and increasing SOD was significant in the medium dose FDBH-treated group(P<0.05 or 0.01).The ALT was increased obviously in all the FDBH-treated groups(P<0.01).But there was no significant difference of BUN,Cr,TNF-a,IL-1 between FDBH-treated groups and TPG-treated group.3.pathological changes:By observed with light microscope,stained with hematoxylin and eosin(HE),and with periodicacid-Schiff(PAS),we could find that the area of mesangial region inmodel rats was widen,the mesangial matrix was aggregated,theglomerular swelling was augmentation,the glomerular cellularity wasincreased,part of glomerular capillary lumina were narrowing orocclusion,part of renal tubules were atrophy even fibrosis.In high,medium dose FDBH-treated group and TPG-treated group,the glomerular swelling is slight,and the mesangial cellproliferative and mesangial matrix was were relieved,and the capillarylumina had no narrowing or occlusion.There was no change in normalgroup.Conclusion:It was suggested that FDBH could decrease the urinary protein of the MsPGN rats effectively,and this has a relation to the effect of improving lipid metabolism,protecting renal function,lessen the injury of renal glomeruli and promoting renal tissues regeneration. FDBH has a better therapeutic effect in MsPGN rats.And it's effect of inhibiting lipid peroxidation,decreasing free radicals,inhibiting the synthesis of TNF-a and IL-1 may be the important mechanism to cure MsPGN.There is a similar effect of dcreasing urinary protein, protecting renal function and nhibiting the synthesis of TNF-a and IL-1 in high,medium dose FDBH-treated group and TPG-treated group.And the effect of decreasing blood-lipids and antioxidation in high,medium dose FDBH-treated group is better than TPG-treated group. There are no changes of hepatic function impairment for long term use.
