Objective Detemining the expression of 5-LOX and LTB4. Analyze the expression of 5-LOX and LTB4 in hepatocellular carcinoma and their relationship with clinical date and prognosis. To elucidate the mechanisms for early diagnosis and clinical application.Methods The specimens were taken from 60 patients with hepatocellulur carcinoma in An Hui Medical Afflicated Hospital from junuary 2006 to December 2007.Record the clicical and follow-up date in detail. The expression of 5-LOX and LTB4 was assayed by Streptavidin-peroxidase immunohistochemistry in 60 hepatocellular carcinomas and adjacent non-cancerous tissues,10 normal liver tissue as control. Experimental date were analyzed together with the clinical pathological datum.Results The expression of 5-LOX and LTB4 in hepatocellular carcinomas was significantly than adjacent non-cancerous tissues and nomal control tissues,and there was significant difference between adjacent non-cancerous tissues and nomal control tissues.positive correlation were found between the expression of 5-LOX and LTB4 in hepatocellular carcinomas and adjacent non-cancerous tissues. Significant differences of the expression of 5-LOX were observed in age and in liver with or without liver cirrhosis in hepatocellular carcinomas.The expression of 5-LOX was higher in younger(<60 years) than in older (≥60 years)(P =0.001); the expression of 5-LOX was higher in liver with liver cirrhosis than without liver cirrhosis(P =0.008). Significant differences of the expression of 5-LOX were observed in tumor number and in liver with or without liver cirrhosis in adjacent non-cancerous tissues. The expression of 5-LOX was higher in multiple tumor than in solitary tumor(P =0.023); the expression of 5-LOX was higher in liver with liver cirrhosis than without liver cirrhosis(P =0.010). Significant differences of the expression of LTB4 were observed in liver with or without liver cirrhosis in hepatocellular carcinomas. The expression of LTB4 was higher in liver with liver cirrhosis than without liver cirrhosis(P =0.003). Significant differences of the expression of 5-LOX were also observed in tumor number in adjacent non-cancerous tissues. The expression of LTB4 was higher in multiple tumor than in solitary tumor(P =0.031). Tumor recurrence rate in one year in patients with the expression of 5-LOX or LTB4 was equivalency compared with whom without the expression of 5-LOX or LTB4 in hepatocellular carcinomas. Tumor recurrence rate in one year in patients with the expression of 5-LOX was higher than whom without the expression of 5-LOX both in hepatocellular carcinomas and adjacent non-cancerous tissues(P =0.044),but Tumor recurrence rate in one year was no significant difference between patients with the expression of LTB4 and whom without the expression of LTB4 both in hepatocellular carcinomas and adjacent non-cancerous tissues.Conclusion 5-LOX and LTB4 play a important role in the formation of hepatocellular carcinoma and possibly have relationships with liver cirrhosis.The expression of 5-LOX is related with tumor recurrence rate in one year and affect the progensis of patients.
