Study On Screening Of Sexual Improvement Chemical Compounds In Chinese Chives And It's Efficacy Idenfication

Chinese chives(Allium tuberosum Rottler.ex Spreng) is perennial herbs belong to the Liliaceae.Its plant and seed are belived having sexual improvement function,but its improving sexual function is still not clear and what compounds in Chinese chives play this role is quite mystery. For confirmation of this ancient legend and found out the sexual improvement compounds the testosterone and NO in serum of mice were measured,the weight of sexual organs of mices was detected and the sex behaviour of mice was observed.The testosterone and NO concentration in serum of mice as indicator for screening some improving sexual compounds from Chinese chives was builted.1,The preliminary experiment of Chinese chives confirm sexual improvement functionThe sexual improvement function experiments of Chinese chives comparing with herba epimedii,herba cynomorii and radices morindae which were acknowledged as chinese herbals having sexual improvement function were carried out.The result was showed that the Chinese chives was confirmed having sexual improvement function,though its efficacy was less than herba epimedii,herba cynomorii and radices morindae. The concents of testosterone and NO of the mouses gavaged with high dosages of Chinese chive extractiong were not significantly more than that of giving middle dosages.This indicated that Chinese chives might be more safe than traditional chinese herbals of sexual improvement function.2,The screening of sexual improvement function chemical substances of Chinese chivesThe crude extract was leached to four phases by petroleum ether, ethyl acetate,n-butanol and water.By doing animal tests to detect the contents of testosterone and NO in serum of mices,the result was showed that the extracts of petroleum ether and n-butanol have sexual improvement function,especial the n-butanol phase.The chemical constituent in the n-butanol extract of Chinese chives was isolated by D101 macroporous adsorption resin eluted with water, 25%,50%,75%and 95%ethanol to obtain the components with different polarity.The fraction of 50%ethanol which has sexual improvement function,isolated by sephadex LH-20,Pre-HPLC to obtain compounds 2,4-Pyrimidinediol(Ⅰ) and 2,4-Dihydroxy-5-methylpyrimidi-ne(Ⅱ).The result was showed that the effects of this two compounds were not significant,because of the losing of separation process.The fraction of 70%ethanol was isolated by RP-18,Pre-HPLC to obtain compound 1-(2-Deoxy-β-D-ribofuranosyl)-5-methyluracil(Ⅲ) and 9-β-D-Rbofuranosyladenine (Ⅳ).The result was showed that the effects of this two compounds were significant,especial 9-β-D-Rbofuranosyladenine(Ⅳ).The chemical constituents in the petroleum ether extract of Chinese chives were separated by silica gel column to obtain different components. The fractions of PE-4 and PE-5 which had this sexual improvement function identified by doing animal experiment were isolated by repeated chromatography on silica gel.The fractions of PE-4-2 and PE-5-2 were identified that both of them had the sexual improvement function.The result was found that PE-4-2 has some substance as anhydride,esters and ethers and PE-5-2 has some substance as ethers and aldehydes which detected by GC-MS.But the experiment was stoped as the limited time.3.Identification of monomers in n-butanol extract of Chinese chivesLC-MS and NMR were used to indentify the monomer's structure. CompoundⅠand compoundⅡwere indentified Uracil and Thymine; CompoundⅢand compoundⅣwere indentified Thymine deoxyriboside and Adenine riboside.Four monomers are all nucleosides which have many physiological functions but their sexual improvement function were not reported.The result was found that Adenine riboside had remarkable effect on improving sexual function,and the effect of gavaged with mixture of four nucleosides was more marked than the effect of monomer. Moreover,substances in the petroleum ether phase also have effect on improving sexual function.It was indicated that the mechanism of Chinese chives's effect on improving sexual function was not only action of single compound but several compounds.Its mechanism may be a specific nutritive action but not a medical action.The results of present study will be useful to further research of chemical constituents and mechanism on Chinese chives.
