This paper applies polyanion-polymer/detergent(PPD) assay for quantifying the high density lipoprotein cholesterol(HDL-C).The formula of reagent I was developed according to the characters of lipoprotein.The interaction of Surfactants with high density lipoprotein(HDL) and low density lipoprotein(LDL) was investigated.The aim of the study was to investigate a new analytical protocol to directly measure HDL-C without precipitation.The kit had low response to LDL when the reagent I was composed of 0.011g/Lα-SCD,1.2g/L Mg2+,0.7 g/L DS and 0.15g/L TOOS;Non-ionic surfactants whose HLB are between 13 and 14 were found to break lipoprotein more effectively with a large number of experiments,the selectivity was lowered when the surfactants were mixed.B-66 showed the highest selectivity to LDL and NP-7 showed the highest selectivity to HDL.At last, when the prepared kit assayed different serotypes which had low,middle and high concentration of HDL,it showed good results on the serum with middle concentration of HDL,however there were some errors in the assayed results of the serum with the low and high concentration of HDL.
