Objective:Discuss the influence of varying degrees and different times of sleep induced hypoxia to emphysema rats model in systemic inflammation and coagulation.Method:50 Wistar rats were randomly and averagely divided into 5 groups,10 each. Group A the rats were breed normally for 28 days,then exposed to the mixed gas of 12.5%oxygen for 3 hours during sleeping time every day;Group B the rats were exposed to cigarette smoke two times every day,about 30 min each time till the end of the experiment;Group C the exposure condition of Group B for 28 days,then still exposed to cigarette smoke and exposed to the mixed gas of 15%oxygen for 3 hours during sleeping time every day.Group D and Group E the same with group C,but exposed to the mixed gas of 12.5%oxygen for 1.5 hours and exposed to the mixed gas of 12.5%oxygen for 3 hours respectively.All the rats were exposed for 56 days continuously,and then killed.Observed pathology of lungs,liver,pancreas and carotid artery.The neutrophils,lymphocytes and alveolar macrophages of the bronchoalveolar lavage fluid were counted.The level of SOD and MDA in liver were determinated.And tested the levels of AT-Ⅲ,FIB,vWF and FVⅢ.Result:(1)Group B,C,D and E got the histopathological changes of emphysema.(2) The total number of cells and lymphocyte ratio of B((5±0.189)×108/L, (6.51±0.547)%),C((5.09±0.178)×10~8/L,(6.68±0.48)%),D((5.18±0.183)×10~8/L,(6.81±0 .637)%),E(5.62±0.266)×10~8/L,(8.51±0.412)%) were higher than that of A((1.64±0.135)×10~8/L,(3.81±0.307)%);and E was higher than B,C and D(P<0.05). (3) MLI of B((91.3±2.261)μm),C((92.8±3.65)μm),D((99.32±2.813)μm) and E((99.32±2.813)μm) were higher than A((46.79±3.182)μm);E was higher than B,C and D(P<0.05).(4)The inflammation score in lungs,liver and pancreas of B((52±5.164)%,(56±4.619)%,(46.5±4.743)%),C((64.6±3.95)%,(65.6±5.06)%,(67±4.2 16)%)),D((61.9±4.254)%,(64.8±5.266)%,(59±6.146)%) and E((73.3±3.86)%, (78±6.325)%,(72.5±4.859)%) were higher than that of A((16.2±2.53)%, (11.6±2.951)%,(13.5±3.375)%);C,D and E were higher than B;E was higher than C and D(P<0.05).(5)SOD vitality of B((76.08±1.764)U/mg),C((67.56±1.847)U/mg)), D((72.24±2.076)U/mg) and E((66.48±1.769) U/mg) were lower than that of A((79.23±2.153) U/mg);C,D and E were lower than B;E was lower than D(P<0.05). MDA content of B((3.01±0.524)nmol/mg),C((3.95±0.378)nmol/mg),D((3.42±0.972) nmol/mg ) and E((6.33±2.363) nmol/mg ) were higher than that of A((1.34±0.079) nmol/mg ),E was higher than B,C and D(P<0.05).(6)The level of AT-Ⅲof B((107.39±8.045)%),C((101.8±4.278)%),D((103.72±6.31)%)and E((99.04±3.986)%)were lower than that of A((116.2±2.53)%);E was lower than B(P<0.05).The level of FIB of B((177.46±7.248)mg/dL),C((182.7±2.776)mg/dL), D((189.98±5.285)mg/dL) and E((198.4±7.369)mg/dL) were higher,than that of A((151.55±6.279) mg/dL);D and E were higher than B;E was higher than C and D (P<0.05).The level of FⅧof B((227.78±10.378)%),C((242.53±14.501)%), D((232.26±4.173)%),E((303.25±32.932)%) were higher than that of A((200.36±4.875)%);E was higher than B,C and D(P<0.05).The level of vWF of B((64.45±1.72)%),C((66.21±3.21)%),D((70.2±8.667)%) and E((71.96±1.851)%) were higher than that of A((56.02±1.465)%);D and E were higher than B;E was higher than C(P<0.05).Conclusion:(1) Severe,prolonged sleep-induced hypoxemia can increase the degree of emphysema;(2)Mild- prolonged and severe short-term sleep-induced hypoxemia can increase the pulmonary and systemic inflammation;severe,prolonged sleep-induced hypoxemia can make more severe systemic inflammation,which based on lymphocytic infiltration.(3)Mild- prolonged and severe short-term sleep-induced hypoxemia emphysema can attenuate antioxidant capacity,severe prolonged sleep-induced hypoxemia can make antioxidant capacity decreased more significantly and only severe,prolonged sleep-induced hypoxemia can increase the oxidation products;(4) Sleep-induced hypoxemia in rats with emphysema aggravate coagulation/anticoagulation system imbalance;Sleep-induced hypoxemia of different degrees and times has different influence on index of blood coagulation and anticoagulation.
