Effects Of Da Huang And Zhe Chong On The Concentration Of Progesterone And Progesterone Receptor In Establishing Animal Models Of The Uterine Leiomyomas

Objective:To study the effects of Da Huang and Zhe Chong on the concentration of progesterone and progesterone receptor in the experimental rats with uterine leiomyomas by loading the rats with estrogen and progesterone.To reveal the curative effect of Da Huang and Zhe Chong on the best proportion of blending,the relation of quantity and effect, the principle of function.In order to improve the curative effect on uterine leiomyomas by Chinese medicine,withdraw the effective component,to lay the foundation of obtain the monomer.Methods:72 healthy female Sprague-Dawley rats were randomly divided into 7 groups:control contrasting group (A),modle group (B),lower dosage group of Da Huang and Zhe Chong (C), medium dosage group of Da Huang and Zhe Chong (D),higher dosage group of Da Huang and Zhe Chong (E),Da Huang Zhe Chong Wan group (F),Gui Zhi Fu Ling Capsula group (G),12 rats in (B) group,10 rats in the other groups.(A) group was treated with 0.2mg peanut oil which is been antiseptic by the high pressure three times a week (Monday,Wednesday,and Friday) for 20 weeks by intramuscular injection.From the 11th weeks on,it was fed with 2ml distilled water six times a week for 10 weeks.the other groups were treated with 0.1mg estradiol benzoate three times a week (Monday, Wednesday, and Friday) for 20 weeks by intramuscular injection. From the 14th weeks on, 0.5mg progesterone (P) was injected intramuscularly once a week for the last 7 weeks.From the 10th weeks on ,2 rats in (B) group were randomly killed to pre-experiment.From the 11th weeks on ,(B) group was fed with 2ml distilled water,(C),(D) and (E) groups was fed with 2ml solution which has Da Huang and Zhe Chong about 0.3g/ml,0.62g/ml and 1.25g/ml,(F) group was fed with 2ml solution which has Da Huang Zhe Chong Wan about0.08g/ml,(G) group was fed with 2ml solution which has Gui Zhi Fu Ling Capsula about 0.04g/ml six times a week for 10 weeks.All rats were killed at the end of 20th weeks.The serum concentrations of progesterone were measured by radioimmunoassay (RIA), Expressions of progesterone receptor in smooth muscle cells were detected by immunohistochemistry.Results:1.The weight of rats with estrogen and progesterone have dropped day after day,and have not rise by Da Huang and Zhe Chong.2.Compared with the control group, the quota of Uterine in modle group was changed significantly(P<0.01).After treated by medicine,weight of uterines were dropped obviously except (C)(P<0.05);uterus index were reduced obviously except (C) and (E)(P<0.05); lengths of left cornua uteri were lessened significantly except (C),(D) and (E)(P<0.01); lengths of right cornua uteri, lengths of cervix uteri and width above cervix uteri were lessened significantly(P<0.01),(C) was the longest of all (P<0.05) ;diameters of cornua uteri were lessened significantly(P<0.01), (C) and (E) were shorter than others (P<0.05); width below cervix uteri were lessened significantly (P<0.01).3.Compared with the control group, the hemorheology of the quotas in modle group was increased significantly(P<0.01). After treated by medicine,blood viscosity was dropped except (C)(P<0.01),(E) was the highest of all (P<0.05),plasma viscosity was dropped except (C) and (E); hematocrit was dropped significantly except (C)(P<0.01),(E) was the highest of all (P<0.05);erythrocyte aggregation index was dropped significantly(P<0.01), (C) was the highest of all(P<0.05).4.Compared with the control group,the concentration of progesterone in modle group was increased obviously(P<0.01),After treated by (D),it was dropped (P<0.05),also (D) and (E) (P<0.01).Compared with (F) and (G),(D) was dropped insignificantly(P>0.05),it indicated that (D) is the same as (F) and (G) in dropping the concentration of progesterone.5.The myometrium of treated rats showed the proliferative features, both multiple and focal.The myometrium were thickened and the smooth muscle cells enlarged.Binuclei and the nucleiin mitotic phase could be easily observed in the cells.And more connective materials constructed the extracellular matrix.All these mentioned tendency could be characterized by excessively focal proliferation,which was similar to those of patients with uterine leiomyoma. 6.The rare PR positive cells were weakly identified in the section of normal uterus. However,the expression of PR positive cells were up-regulated distinctively in hormone treated rats.(C),(D),(E),(F) and (G) presented to decrease the positive cell density significantly vs model group (P<0.01).Compared with (F),(C) was dropped significantly(P<0.01), Compared with (F),(C) was dropped (P<0.05), Compared with (F),(G),(D) and (E) was dropped insignificantly(P>0.05),it indicated that (D) and (E) is the same as (F) and (G) in decreasing the positive cell density.Conclution:1.Animal Models of the Uterine Leiomyomas were established successfully by Loading the Rats with Estrogen and Progesterone.2.Da Huang and Zhe Chong can reduce the weight of uterines,uterus index,lengths of left and right cornua uteri,lengths of cervix uteri,width above and below cervix uteri of rats'Uterine with Estrogen and Progesterone obviously.3.Blood stasis was established in the experimental rats with uterine leiomyomas through the hemorheology of the quotas.4.The proliferative features was similar to those of patients with uterine leiomyoma.5.The concentration of Progesterone and Progesterone Receptor play a important part in the growth of Uterine Leiomyomas,Da Huang and Zhe Chong,which was demonstrated to reduce the concentration of progesterone and suppress the expression of progesterone receptor ,may be one of reasons to treat Uterine Leiomyomas.
