Study Of Cell Perfusion-NMR

To explore mechanism of medicine therapy tumor cells and select medicine efficacious ingredient,in this paper,a method of NMR-cell perfusion was established in order to monitor the process under drug stimulation.NMR is a technique,which continuously observes energy metabolism change in lively tissues and cells without damage,but it requires larger numbers and longer survival time of cells compared to bio-chemical methods.Therefore,it is necessary to design a set of cell perfusion device which meets NMR test with higher cell density and longer cell living time.In this paper,a set of cell perfusion-culture device was designed and its performance tested.Under normol perfusion conditions,experiments showed no germ grew,and temperature,pressure,flow rate of the device were relatively invariable.Perfusion-culture of breast cancer cells in the device indicated that cells could survive for more than a hundred hours. through the cell-perfusion-NMR technolgy observed metabolism changes of cantharidine action on breast cancer cell.Experiments shown the method of NMR cell-perfusion can observe changes of energy metabolism in cells,can confirme efficacious ingredients in drugs and deduce acting time of drugs to tumor cells.The cell-perfusion-NMR online observation apparatus,which can be used for study on medicine reacting with tumor cells,provides a scientific basis for anti-tumor mechanism of traditional Chinese medicine.The NMR-cell perfusion method,founded in the work,is promising to become a new method for research on medicine efficacious ingredients.
