ObjectiveFumonisins(FB) are a group of mycotoxins mainly produced in maize and maize-based products by F.moniliforme and F.proliferatum.They can effect humans and animals which associated to esophageal cancer.FB1 is known to be the cause of equine leukoencephalomalacia and porcine pulmonary edema syndrome.Available epidemiological evidence has suggested a link between dietary fumonisin exposure and human oesophageal cancer in some locations with high disease rates.As the FB pollution often occurs in the warm areas of the world,our native Country investigated the FB1,which mostly concentrate on south regions,but not the northward.Our study aimed at developing a liquid chromatography/tandem mass spectrometry method,and test the FB1 in maize comparing the influences of different seasons and different climate conditions.For the sake of depressing oesophageal cancer rates in KangPing,then offering basic data for establishing FB1 standard in corns, protecting human's health and promoting international trade.MethodGrind corn laboratory sample to yield material of such size that can 90%is retained between 250μm-500μm mesh screens.The ground sample(1 g)was weighted into a 50 ml polypropylene centrifuged tube and extracted with 10 ml of extraction solvent acetonitrile:water:acetic acid(49:50:1,V:V:V) by shaking for 30 min and was then centrifuged for 10 min at 4000 rpm,1 ml of the extract was transferred into another 10 ml glass tube and evaporated to dryness at 50→C under a stream of air.The residue was reconstituted in 1 ml of the mobile phase and vortexed for 30 s,and the solution filtered through a 0.22μm syringe filter.20μl of the final solution was injected into the LC-MS/MS system. The analyte was extracted from maize samples by acetonitrile:water:acetic acid (49+50+1,V:V:V),raw extracts were evaporated to dryness and dissolved with mobile phase,chromatographyed on a C18 column and detection was performed on a triple quadrupole tandem mass spectrometry with electrospray ionization source.ResultsThe method has a limit of quantification of 10μg/kg in maize samples.The calibration curve was linear in the range of 1-500 ng/ml.The mean recovery ranged from 94%to 104%and the relative standard derivations were lower than 9%.ConclusionThis method was sensitive and specific to be applied for the determination of fumonisin B1 in maize samples.
