introductionTumorous bionomics depends on not only the intranuclear genetic material,but also the extranuclear mitochondrial DNA.MtDNA has higher mutation rate because of variant reasons and may have potential carcinogenicity.The mutational mtDNA can make continued colonization of the human genome just like DNA virus or RNA virus and induce the malignant transformation of normal cells.MtTFA is synthesized in cytoplasm,then transfened into mitochondrium and controls the synthesis of mitochondrial respiratory chain components.Human mitochondrial DNA is packaged with mtTFA.Mitochondrial transcription factor A is a key regulator involved in mitochondrial DNA transcription and replication.The recent studies show up-regulation of mtTFA in rapidly growing tumors perhaps regulate the expression of gene involved in tumorigenesis and tumor proliferation.MtTFA can recognize and bind damaged mtDNA and significantly impede recognition and repair of mtDNA adducts by repair enzyme,followed by tumorigenesis.The interaction of p53 with mtTFA may play an important role in apoptosis.This experiment was performed to observe the expression of mtTFA and elucidate the association with arising and development of cervical cancer.Materials and methods1.Reverse transcription polymerase chain reaction(RT-PCR)Sample:the cervical cancer group(30 cases in total) and the normal group(30 cases) were obtained from the department of gynecology and obstetrics of shengjing hospital affiliated to China Medical University.All patients had no radiotherapy or chemotherapy before operation and all samples were confirmed by pathology department after operation.RNA extraction:RNA were Extracted by using RNA extraction kit. cDNA synthesis:cDNA synthesis was performed by using Reverse transcription kit.The PCR reaction consisted of an initial denaturation step(94℃,3mins),followed by 35 cycles of denaturation(94℃,35s),annealing(49℃,1min),and extension(72℃, 1 min).A final Extension step(72℃,10min) terminated the reaction.Amplification products were then analyzed by electrophoresis in a Gene Finder-stained 1.5%agarose gel at 100V for 30min,and visualized under UV light.2.Statistical analysis:The software SPSS16.0 was applied for statistical analysis.T test and ANOVA variance analysis were used to compare cases and controls.Results1.The expression of mtTFA in cervical cancer and normal cervix tissuesThe expression of mtTFA in cervical cancer and normal cervix tissues were 0.28±0.10 and 0.19±0.13,respectively,with statistical significant difference(P<0.05).2.To compare the correlation of the expression of mtTFA and clinical parametersThe expression of mtTFA was associated with FIGO stage(P<0.05),but not associated with other parameters(P<0.05).Conclusion1.The levels of mtTFA expression of cervical cancer group were significantly higher than those of normal group.(P<0.05).2.The expression of mtTFA was associated with FIGO stage(P<0.05),but not associated with other parameters(P>0.05).3.The abnormal expression of mtTFA in cervix might be involved in the the arising and development of cervical cancer.
