Objective1.To investigate the relationship between the birth weight of fetus and the activities of P70S6K.2.To investigate whether insulin and rapamycin can affect the activities of P70S6K by the mTOR/P70S6K signaling pathway in the cells of the placental tissues.Methods70 cases of mature placental tissues after uterine-incision delivery without complications(such as hypertension,diabetes,heart disease,endocrine disturbance,fat metabolism abnormality) in the gestational period were selected as the investigated subjects,body mass index of the progestation is normal(20≤BMI≤24.0),body weight gain in the duration of pregnancy is alike.(except abnormal placenta and umbilical)To investigate the relationship between the birth weight of fetus and the activities of P70S6K.they were divided into three groups:①group1(the weigh of the fetus≥4000g)②group2(3000g≤the weight of the fetus<4000g)③group3(the weight of the fetus<3000g) according to their birth weights,there are 20 cases in the every groupTo investigate whether insulin and rapamycin can affect the activities of P70S6K by the mTOR/P70S6K signaling pathway in the cells of the placental tissues that were divided into three group according to stimulating factors. ①insulin group②rapamycin group③control group There are 10 cases in the every group。Activities of P70S6K were carried out by immunoprecipitation,incoparation of [r-32P]ATP,tissue cell culture.Results1.There were significant statistical differences in the comparison of P70S6K activities,group1>group2>group3,(group1,2:P<0.05,group2,3:P<0.05 group 1,3: P<0.01).2.The P70S6K activities in the placenta from the insulin stimulation group was significantly elevated when compared with that of the control group,and the statistical significance was found in the difference(P<0.01);P70S6K activities in the placenta in the rapamycin group and it was statistical significant when compared with the insulin group(P<0.01).Conclusion1.The activities of P70S6K protein in the placental tissues increased with the increase in the birth weight of the fetus and the activities of P70S6K was also regulated by the mTOR/P70S6K signaling pathway,indicating that there was certain relationship between the mTOR/P70S6K signaling pathway in the placental tissues and the birth weight of the fetus.2.Insulin and rapamycin can regulate the activities of P70S6K in the placenta,indi-cating that insulin and rapamycin can regulate the activities of P70S6K in the placenta by the mTOR/P70S6K signaling transduction pathway and insulin regulate birth weight completely or partly and further affect the birth weight of the fetus.
