IntroductionArticular cartilage,because of its lack of its own blood circulation system,only to provide the majority of joint synovial fluid nutrition,it will be difficult to repair when it is damaged.Cartilage tissue engineering in the current study,autologous chondrocytes as seed cells are facing the problem:in vitro cartilage cell growth slow to happen easily differentiated,so that access to meet the tissue-engineered cartilage to build the necessary and sufficient quantity to maintain its normal Seed phenotype of cartilage cells.So how in the use of appropriate concentrations of growth factor cytokines,such as the United stimulated chondrocytes through appropriate extension of the body on behalf of the number of rumors,there is enough in favor of access to both quantity and maintain a good chondrocyte phenotype,so that as autologous chondrocyte cartilage seed cells in tissue engineering has become a hot research.BMP-14 is a multifunctional growth factor,which is one of important regulatory factors of the body growth and development,it is an important role on induction of cartilage in the formation of bone, cartilage,tendon repair ligament damage.FGF stimulate proliferation and differentiation of cartilage cells,resulting in mitogen-shape formation.It is the strongest currently known to promote cell growth factor.ObjectiveThe purpose of this study is to investigate the effects of BMP-14 and bFGF,used in combination or alone,on proliferation in rat articular chondrocytes in vitro.To find the best combination and best concentrations of different growth factors on proliferation of chondrocytes in cartilage tissue engineering.MethodsArticular chondrocytes of Wistar rats were isolated and cultured in-vitro.Make the second generation of chondrocyte cell which is at good function as the experimental distribution after the add different combinations and concentrations of growth factor.Sixteen study groups were set,and each group was cultured in different concentrations of BMP-14 and/or bFGF.In the experiment,the concentrations of BMP-14 were 0ng/ml,50ng/ml,100ng/ml,200ng/ml and the concentrations of bFGF were 0ng/ml,5ng/ml,10ng/ml,20ng/ml.The cell numbers were counted 0,2,4 and 6 days after growth factors were given for comparing the effects of BMP-14 and bFGF on proliferation.The expression of type-â…¡collagen was investigated immunohistochemically with image software,and the expression of proteoglycan was studied using Alcian blue staining.ResultsBMP-14 can stimulate neither proliferation of chondrocyes,nor expression of type-â…¡collagen or proteoglycans.While bFGF has an positive effect on both cell proliferation and cytokine expression.BMP-14 and bFGF showed an synergic effect when used in combination.ConclusionThe study suggests that the combination of BMP-14 and bFGF may be an applicable method on proliferation in rat articular chondrocytes for in the cell culture of cartilage tissue engineering.
