Zedoary Yellow On Rectal Cancer Cells In Vitro

Objective:To probe into the pharmacological influence of Chinese medicine E Huang Anti-tumor Prescription(EHAP) on proliferation and apoptosis of human CoLo320 cell in vitro,study on the biological basis of its treatment of rectal cancer,in an effort to find new medicine and provide theoretical basis to prevent rectal cancerMethod:Colo320 cells were isolated and purified from human rectum. After activated by beingcultured in vitro.The influence of 40%, 20%,10%,5%,2.5%,1.25%,six different concentration of EAHP groups on the Colo320 cells were separately compared with control group, whose serum not containing drug.The influence on Colo320 cells proliferation was observed by means of growth curve and MTT assay; and flow cytometry(FCM) was used to determine the apoptosis and cell cycle after the treatment of EHAP.To show the relationship between the action of EHAP and the apoptosis rate of Colo320 cells.Result:1.MTT showed cell proliferation was inhibited after 24h action of EHAP,all the inhibitory rates were remarkably higher while compared with the control group separately(P< 0.01) The 10%group was the highest,then the 5%group,and the 1.25%group got the lowest2.FCM analysis showed that G 0/G 1 and G 1/S phase rate increased while S and M /G2 phase rate decreased after Colo320 cells were exposed to EHAP for 24 h3.Typical apoptotic peak appeared after treatment with EHAP for 48h,apoptotic rates were obviously higher than the control group with significant difference(P<0.01).The 20%group was the highest,the 1.25%group got the lowest.Conelution:EHAP influences cell cycle,inhibits proliferation,and induces apoptosis in human Colo320 cell to anti-tumor.
