Objective:The treatment mechanism of Holographic Meridian Scraping Therapy(Guasha) has yet to be studied and explained,which is an important means of disease treatment and prevention in Traditional Chinese Medicine clinically or among the Chinese folks. We have brought out a hypothesis that Guasha is capable of activation of heme oxygenase-1,catalytic degradation of heme,yielding powerfully protective and beneficial products,such as bilirubin;activation of superoxide dismutase(SOD); IL-regulation(including IL-1,IL-6) and interleukin,etc.And related research,to some extent,from the perspective of modern science,reveals the mechanism of Guasha as a safe and effective Chinese medicine therapy in order to further advance the clinical development,provide basis and reference for efficacy assessment and further study of the mechanism of Guasha.Research methods:1 Theoretical ResearchBy sorting out literature and logically analyzing,we get to know the origin, history,evolution of Guasha,form a theoretical basis and put forward the therapy may have anti-oxidation and regulation of immune function.2 Experimental Research2.1 Experimental research methods in animalsmale Wistar rats are randomly divided into A,B,C,D four groups,10 in each group and marked in number.Blood is drawn from the rats' heart in Group A and kept in cold storage at 4℃.Rats in Group B,C and D are shaved at the back and then skin scraped along the Du Meridian at the back from head to tail with a skin-Guasha plate made of cattle horn for 5 minutes at 70 times/min.Some Sha points(toxin) appear on the back of rats like that of humans.Blood is drawn from the rats' heart 0.5h in Group B,6h in Group C and 12h in Group D after being skin scraped.All the blood samples are marked "kept at 4℃in cold storage for next day's test.The blood samples are tested blood routine,IL-1 and bilirubin respectively in accordance with the reagent kit manual.SOD was determined by xanthine oxidase in accordance with SOD kit manual to make detection reagent.2.2 Research methods of test on humanTestees are students of 21-24 years old,8 males and 8 females without any discomfort,having uniform diet during the whole process.Two groups of upper-arm venous blood are collected 1ml in EDTA tube and 2ml in general blood tube in clinic. The testees in sitting prone position are skin scraped with skin-Guasha plate made of cattle horn along the entire back from top to bottom down the Du Meridian for 5 minutes at 70 times/min.Upper-arm venous blood is drawn 0.5h and 6h respectively after Guasha.The blood samples are tested blood routine,IL-1,IL-6 and bilirubin respectively in accordance with the reagent kit manual.SOD was determined by xanthine oxidase in accordance with SOD kit manual to make detection reagent.2.3 The statistical software of SPSS10.0 is applied to analyze the experimental data, to calculate mean±standard deviation,and when P<0.05,it is statistically meaningful according to one-factor analysis of variance,q-text and t-text.Results:1 Results of theoretical ResearchBy sorting out literature and logically analyzing,we get to know the origin, history,evolution of Guasha,form a theoretical basis and put forward the therapy may have anti-oxidation and regulation of immune function.2 Results of exeperimental research2.1 The impact on antioxidation function of organism by Guasha2.1.1 Direct bilirubin experimental data in rats shows that there is no difference between Group A and Group B(P>0.05);There is s significant difference between Group A and C,Group A and D(P<0.05);There is a difference between Group and C, B and D(P<0.05).There is no significant difference between Group A and Group B or Group C in data of total bilirubin(P>0.05);There is significant difference between Group A and Group D(P<0.05);There is significant difference between Group B and Group D,Group C and Group D(P<0.05).There is no significant difference between Group A and Group B or Group C in data of SOD(P>0.05); Group A is greatly lower than Group D(P<0.05).2.1.2Direct bilirubin and total bilirubin in the female group of human trials significantly increase 0.5-6 hours after being Scraped(P<0.05);Measured value for male direct bilirubin and total bilirubin significantly decrease 6 hours after being Scraped(P<0.05).SOD for female group has increased somewhat post-Guasha,but it has no meaning in statistics due to P>0.05.SOD for male group has increased greatly 0.5-6 hours after being Scraped and it is meaningful in statistics due to P<0.05.2.2 The impact on immune function of organism by Guasha2.2.1 Experimental data of Lymphocytes in rats shows that there is no significant difference between Group A and Group B or Group D(P>0.05);Group A and Group C show a downward trend and significant difference;There is significant difference and upward trend between Group B and Group D(P<0.05);Group C and Group D show upward trend and significant difference(P<0.05).Experimental data of Leucocytes in rats shows that there is no significant difference between Group A and Group B or Group D(P>0.05);Group A and Group C show significant difference (P<0.05);There is difference between Group C and Group B or Group D(P<0.05). Granulocyte data in the C group and D group decreased than that in group A,but there is no significant difference between any two groups(P>0.05).Rat IL-1 data show a slight downward trend 0.5 hours after scrape,however,there is no statistical difference between different group of rats(P>0.05).IL-6 data has unstable results maybe due to small amount of samples while significantly different values.2.2.2 After scrape,for leucocytes of male groups and female groups of Human trials and lymphocytes of female group,there is a significant difference between A group,B group and C group(P<0.05).Lymphocytes of male group increase greatly 0.5-6 hours after scrape(P<0.05).The absolute value of neutrophils for male group and female group changes little after scrape.IL-1 of female group increase somewhat 6 hours after scrape,while male group decreases somewhat;IL-6 for male group and female group increase somewhat 0.5-6 hours after scrape,but statistically meaningless due to P>0.05.Conclusion:1 By studying theories of Guasha and its origin,history,changes and the theoretical foundation,we know Guasha has a long history,unique in its approach, simple and secure,adapted to a wide range of situation reliable efficacy and greatly welcomed by the broad masses of the people.It has the role of promoting blood circulation by removing blood stasis,enhancing blood flow,promoting the circulating of Qi,promoting the circulating of meridians and collaterals,killing pain,eliminating evil and toxins,clearing away heat and toxic material,inducing resuscitation and refreshment and strengthening the spleen and stomach.Its treatment range has been widely expanded and not limited by its traditional scope,effective in helping treat 11 major categories of more than 400 kinds of diseases in internal medicine,surgery, gynecology,paediatrics and ophthalmology.With the rise of natural therapy,Guasha, as a natural therapy capable of both preserving health and curing diseases,will receive more and more attention and become an integral part of China's health care.And the hypothesis is explicitly put forward that the therapy affects the anti-oxidation and immune function.However,to study the principles of how Guasha prevents and cures diseases in a systematic way,as in the whole awareness of traditional Chinese medicine,there is the lagging and inadequateness,not only limiting the popularization and application of Guasha,but also seriously affected and restricted the study and development of Guasha Therapy.However,modern medical research on the mechanism of Guasha therapy is still less and preclinical and clinical trials and researches should be further studied in order to provide a more adequate theoretical and clinical basis for its own popularization and application.2 The results of human trials show that Sha points(a kind of toxin) will also appear on normal humans to varying degrees after a Guasha,the experimental data measured within the normal range.Therefore the thought that normal humans can also get Guasha,as a means of preventive health care,is also in line with the idea of traditional Chinese medicine,"To treat when illness is not coming".3 Experimental results of the impact on the antioxidation function of the organism by Guasha show that direct bilirubin of rats has an upward trend,most significantly 6 hours and 12 hours after Guasha,hinting that increasing direct bilirubin may be part of the mechanism for Guasha to improve antioxidant ability of organism, decrease the damage on organism by free radical and thus achieving the aim of preventing diseases and preserving health.If Guasha could increase SOD level of organism within normal range,it certainly could improve antioxidant ability of organism,decrease the damage on humans by free radical.The results of human trials show that SOD level of organism changes within normal range after Guasha. Therefore,increased direct bilirubin and SOD may be one of the mechanisms to enhance antioxidant defense system of the organism by Guasha and it may be part of the causes and experimental evidence for Guasha as a safe and effective method of Traditional Chinese Medicine and the civil treatment.4 Experimental results of impact on immune function of organism by Guasha show that Guasha and its "Sha points" phenomenon can stimulate the increase of the interleukin,IL-1 and IL-6 and correspondingly it will promote proliferation and activation of the B cells and T cells,affecting the regulation of immune system of organism,so as to achieve the purpose of resisting and treating diseases.IL-1 and IL-6 in IL has the role of promoting growth,differentiation and proliferation of immunocyte;regulating immune responses and its intensity.The measured values of interleukin and IL-6 in male groups and female groups of Human trials have the upward trend,suggesting that IL-6 in male and female group may be part of the mechanism for scrape to enhance immunity of organism,so as to achieve the role of preventive health care.Thcrefore,Guasha,as an inducement factor,increases white blood cell and IL-6 and stimulates the proliferation and activation of immune cells to enhance immune ability of organism,which may be part of the mechanism for preventive health care.
