The Effects Of Bortezomib And Dexamethasone In The Treatment Of Relapsed Or Refractory Multiple Myeloma

Objective:To evaluate the effects and toxicity of Bortezomib and Dexamethasone in treatment of relapsed or refractory multiple myeloma.Method:18 patients with relapsed or refractory multiple myeloma were treated with Bortezomib and Dexamethasone(Bortezomib 1.0-1.3mg/m2 iv d1,4,8,11;Dexamethasone 40mg d1,2,4,5,8,9,11,12).One cycle is 21 days.All patients were received 1-4 cycles,recorded the curative effect according to the IMWG standard and side effects according to the WHO.Result:After one cycle,1 patient got complete remission(CR),2 patients got very good partial remission(VGPR),and 11 patients got partial remission(PR).The rate of ORR(CR+VGPR+PR) was 78.8%.2 cycles later,of all 9 patients,3 patients got VGPR, and 5 patients got PR.The rate of ORR was 88.9%.3 cycles later,of all 6 patients,4 patients got VGPR,and 1 patients got PR.The rate of ORR was 83.3%.After 4 cycles, of all 5 patients,2 patients get VGPR,and 2 patients got CR.The rate of ORR was 80.0%.Of 3 patients with renal dysfunctional,1 patient got VGPR and the renal function had graet improved,2 got SD and the renal function had not aggraved after the treatment of Bortizomib.The patients who had the chromosome abnormality one got CR and the other got VGPR.Main toxicities were peripheral neuropathy(38.9%), debility(11.1%),thrombocytopenia(33.3%),the infection of herpes zoster virus(16.7%), dysfunction of Hepatic function(16.7%).Conclusion:The effects of Bortezomib and Dexamethasone in treatment of relapsed or refractory multiple myeloma are inspiring.The effects were raising with the accounts of using Bortezomib and Dexamethasone.It is safe for the patients who have the renal dysfunctional and can improved the renal function.The abnormality of chromosome do not effect the effects.Side effects were be tolerance and the regiment was safe.
