Objective:To investigate the expression of Survivn and RASSF1A in colorectal cancer and its clinical significance.Methods:Reverse transcription PCR was used to detect the expression of Survivin and RASSF1A in 60 cases of tumor tissue and 20 cases of surrounding normal colon tissue. Methylation-specific PCR(MSP) was used to investigate the promotor methylation of RASSF1A.Results:We found that Survivin is expressed in 28 cases of tumor tissue but only 1 case of normal tissue,RASSF1A was lost in 18 cases of tumor tissue but no case of normal tissue.We also found 27 promotor methylation in 27 cases of tumor tissue. Expression of Survivn was associated with metastasis,tumor differention and Dukes' classification(P<0.05).Expression of RASSF1A was associated with metastasis, tumor differention and Dukes' classification(P<0.05).Promotor methylation of RASSF1A was associated with tumor differention(P<0.05) and the expression of RASSF1A(P<0.001).Conclusion:Poromotor methylation of RASSF1A leads to absence of RASSF1A expression.Abnormally expression of Survivin and RASSF1A contributes to genesis and development of colorectal cancer.Expression of Survivin and RASSF1A andpromotor methylation of RASSF1A may be served as biomarkers for colorectal cancer diagnosis and prognosis.
