Tibetan medicine is one of the important traditional medicine in China ,and is receiving increasing because of its safety and unique effect. Tibetan Medicine which is called RanNaSangPei(RNSP)is one of Tibetan flavor pearl pills, It is one of the most valuable Tibetan medicine. Experience on behalf of the constant practice of Tibetan medicine and clinical observation in recent years indicate that the drug have better effectiveness on stroke, paralysis, hemiplegia, infantile convulsions, epilepsy, brain disease after surgery recovery, heart disease, hypertension and autonomic disorders, and so on .Clinical and experimental studies have shown that RNSP is effective on the treatment of cardiovascular and cerebrovascular disease, Its mechanism is still not very clear. ObjectiveThis experiment is to observe the effect of Rannasangpei(RNSP) on hemorrheology after the administration of preventive to rats, and the role on lipids level in hyperlipidemia rats. Provide the experimental evidence of pharmacodynamic action and mechanism in order to application in cardiovascular diseases.Methods1. The effects of RNSP on hemorrheology in acute blood stasis model rats.Wistar rats 84, weight between 180-220g, were divided into normal control group, model group, RNSP high dose group (2g/kg), RNSP middle dose group (1 g / kg), RNSP low dose group (0.5g/kg) and Danshen group (2g/kg) randomly. Each group has 14 rats. Every day the rats are given dose or normal saline one time for 10 d. After the administration of preventive, the blood stasis model in rats induced by adrenaline, the parameters of hemorrheology were measured, such as whole blood viscosity (low, medium, high shear), plasma viscosity, hematocrit, whole blood reduced viscosity (low, medium, high shear), erythrocyte sedimentation rate, fibrinogen, erythrocyte sedimentation rate equation k value, erythrocyte aggregation index, erythrocyte rigidity index and erythrocyte electrophoresis index.2. The effects of RNSP on serum lipids level in rats.Wistar rats 84, weight between 180-220g, 70 of the rats were feed by high cholesterol diet 20d ,and divided into model group, RNSP high dose group (2g/kg),RNSP middle dose group (1 g / kg),RNSP low dose group (0.5g/kg) and Xuezhikang group (2g/kg) randomly. Each group has 14 rats. Every day the rats are given dose or normal saline one time for 4 week, and continue to feed to high cholesterol diet. The surplus 14 rats were feeding by normal diet as normal control group. Then we observe the effect of RNSP in rats with hyperlipemia on triglyceride (TG) , total cholesterol(TC), high density lipoprotein cholesterol(HDL-C), and low density lipoprotein cholesterol(LDL-C).Results1. The results suggest that RNSP markedly decreased whole blood viscosity, plasma viscosity ,whole blood reduced viscosity and fibrinogen.At same time it reduced the aggregation index of RBC(AI)and hemotocrit(HCT) and so on, but no statistics significance.The results showed that RNSP medium-dose group on low shear rate whole blood viscosity is (14.848±2.304),RNSP high-dose group on low shear rate whole blood viscosity is (14.598±2.079),RNSP high-dose group on medium shear rate whole blood viscosity is ( 8.608±0.557 )。RNSP high-dose group on plasma viscosity is (1.582±0.152)。RNSP high-dose group on low shear rate whole blood reduced viscosity is (27.876±2.897),RNSP medium-dose group on low shear rate whole blood reduced viscosity is(29.841±3.323),and on medium shear rate whole blood reduced viscosity is(15.527±1.752)。RNSP medium-dose group on fibrinogen is (4.55±0.72),RNSP high-dose group on fibrinogen is(4.45±0.81)。It suggests RNSP high-dose group on whole blood viscosity (low shear, medium shear) compared with the model group has significantly decreased(p<0.05), and RNSP medium-dose group on whole blood viscosity (low shear) compared with the model group has significantly decreased(p<0.05), but on high shear rate whole blood viscosity has little effect ,at same time RNSP high-dose group on plasma viscosity has significantly decreased(p<0.05)。The results also suggested RNSP medium-dose group on whole blood reduced viscosity (low shear) compared with the model group has significantly decreased(p<0.05)。And RNSP high-dose group on whole blood reduced viscosity (medium shear) compared with the model group has significantly decreased(p<0.05), in particular, it has large change on whole blood reduced viscosity (low shear)(p<0.01)。At last RNSP can significantly decreased fibrinogen compared with the model group(p<0.05)。2. The results suggest that TC (1.991±0.415 )mmol?L,TG (1.386±0.259 )mmol?L,HDL-C (0.656±0.207) mmol?L and LDL-C (0.838±0.148) mmol?L in model group were decreased significantly compared with control group. That TC(1.546±0.346) mmol?L,TG(1.011±0.273 mmol?L),HDL-C(0.858±0.181 mmol?L) and LDL-C(0.689±O.144 mmol?L)in Xuezhikang group were decreased significantly compared with the model group(p<0.01). RNSP low-dose, medium-dose and high-dose group whose TC was (2.042±0.377),(2.032±0.459),(2.031±0.267 );TG was (1.264±0.239),(1.319±0.287),(1.318±0.216)and LDL-C was (0.872±0.219),(0.869±0.232),(0.826±O.149)were no significantly differences compared with the model group(p>0.05). In conclusion, Rannasangpei has no significantly effect on serum lipids level in rats. .ConclusionsRNSP has an obvious improvement effect on hemorheology, and can promote the blood circulation to remove blood stasis. But it was observed that the serum lipids level on hyperlipemia rats was not perceptibly changed. And the drug may be change whole blood viscosity of low shear and low shear whole blood reduced viscosity, plasma viscosity, fibrinogen on"blood stasis" in rat, to inhibit erythrocyte aggregation and change of blood stasis model rats "thick, sticky, poly, condensate" state, to improve hemorheology. This is probably the one of mechanisms for the treatment of RNSP cardiovascular and cerebrovascular disease.
