| Background: More and more research prove thatthe gastric bypass operation on type 2 diabetic rats have an significant effection,surgical treatment on diabetes were accepted by more people,but we still do not know the mechanism of the suegical treatment. And we need to explore the ture.Objective:Investigate the mechanism of gastric bypass surgery (Gastric Bypass, GBP Roux-en-Y) to treat type 2 diabetic (non-insulin-dependent diabetes, non-insulin dependent diabetes mellitus, NIDDM) rats which have Normal weight. Methods: 20 type 2 diabetes mellitus animal model in rats induced by diets and alloxan,and divided the type 2 diabetes mellitus animal model into two groups:Type 2 Diabetes-operation group (DO group) and Type 2 Diabetes -control group (DC group);The 8-weeks-age wistar rats were randomly divided into two groups:, Normal-operation group (NO group) and Normal-control group (NC group),record the Blood Glucose and the Gastric inhibitory polypeptide (GIP),the glucagon-like peptide-1 (GLP-1) were measured respectively at fasting and post oral glucose 30min condition.Results: 1.Compared with the preoperative, the fasting blood glucose of DO group rats 1 week post operation has begun to decline, the fasting blood glucose from 23.04±3.51mmol / L decreased to 15.67±1.51mmol / L (P <0.05), 8 weeks post operation the fasting blood glucose decreased to 5.97±0.73mmol / L (P <0.05), and keep the low level for weeks, the fasting blood glucose of DO group was significantly lower than the DC group post operation (P <0.05), the fasting blood glucose of NO group has no significant changes post operation; DO group decreased more obvious post oral glucose 30min condition (P <0.05), NO group has begun to decline post operation two weeks, and decreased to 7.84±0.84mmol / L (P <0.05) eight weeks post operation, DC group and NC group have no significant changes post operation.2. Compared with the preoperative, the fasting GIP of DO group 1 week post operation has begun to decline, the fasting GIP from the 11.14±1.62pmol / L decreased to 6.98±0.91 pmol / L (P <0.05) eight weeks post operation. And the fasting GIP of NO group has begun to decline from 1 week post operation, the fasting GIP from the 12.24±1.43pmol / L decreased to the 7.46±0.74pmol / L (P <0.05) eight weeks post operation ; the fasting GIP of DO group was significantly lower than the DC group post operation (P <0.05) and NO group was significantly lower than the NC group post operation (P <0.05); the fasting GIP of DC group and NC group have no significant changes post operation; And it is more significantly decline on post oral glucose 30min condition.3. Compared with the preoperative, the fasting GLP-1 of DO group 1 week post operation has begun to rise significantly, the fasting GLP-1 from the 6.84±0.84pmol / L increased to 12.64±1.59 pmol / L (P <0.05)1 week post operation, increased to the 23.82±2.07 pmol / L (P <0.05) eight weeks post operation; the fasting GLP-1 of NO group has begun to rise from 1 week post operation, the fasting GLP-1 from the 10.65±1.48 pmol / L rise to the 14.23±1.73 pmol / L (P <0.05) eight weeks post operation; the fasting GLP-1 of DO group was significantly higher than the DC group post operation (P <0.05) and NO group was significantly higher than the NC group post operation (P <0.05); the fasting GLP-1 of DC group and NC group have no significant changes post operation; And it is more significantly rise on post oral glucose 30min condition.Conclusion: The GBP operation on type 2 diabetic rats have an significant effection , it is play an important role about the high secretion of GLP-1 , which caused by food into the last-jejunum directly post operation. |
