Objective To obtain high purity of oligodendrocyte progenitor cells (O2A) ,which get from new born rats cerebrum by culture and purification,and to observe the morphological changes during the course.Then put the O2A cells under the conditions of hypoxia ischemia for different time, to study the morphological changes under this conditions.At the same time immunofluorescence and western blot are done to explore hypoxia ischemia on O2A Tau protein in vivo.Methods O2A were isolated by constant temperature orbital shaking twice and identified by immunofluorescence technique with A2B5 protein,then taken photos with inverted fluorescence microscope camera, and we got its purity.Then we created low-sugar hypoxia environment artificially ,which according with the conditions of ischemia and hypoxia in vivo, so we set the model of ischemia and hypoxia in vitro for O2A,then may find its morphological changes at the various time points, and we got fluorescence intensity of it with laser scanning microscope and used western blot to gain its gray value,which reflects the amount of Tau protein and show its tendency. Results O2A's body is round,with unipolar or bipolar processes,and the cells may get together to form cloning balls.It shows positive identification when tagged by A2B5, and the purity is over 85 percent by immunefluorescence. With the extension of time for ischemia and hypoxia, the quality of O2A cells gradually reduce, and the processes gradually shorten .At the same time,the fluorescence intensity and gray value gradually weakened,which show the amount of Tau protein gradually reduce.Conclusions 1. O2A cells may be cultured and purified in vitro;2. O2A change their forms as the time extends for ischemia and hypoxia.,and most of the cells die for 12 hours and turn to debris.3. With the time of ischemia and hypoxia goes on, the fluorescence intensity gradually weakened,and it shows the amount of Tau protein gradually reduce.
