Salivary Adenoid Cystic Carcinoma is a kind of tumor with high-grade malignance, commonly found in head and neck malignant neoplasms. the patent's prognosis is not good for its high-grade malignance and easy recrudescence after surgery. Therefore, how to predict and prevent such kind of disease effectively has becoming an important topic. Along with the development of molecular biologic technology, more and more genes have been deemed as relating to the occurrence and development of such tumor. By comprehending these genes'functions in this tumor, it can help us to seek for the methods to diagnose and treat the tumor and predict its development tendency.Objective:On the basis of the proved the correlation between Notch-1 of the Notch family and Salivary Adenoid Cystic Carcinoma at the preliminary stage,our research will do further research in the correlation between the other genes of Notch signal pathway and Salivary Adenoid Cystic Carcinoma.Methods: According to the Salivary Adenoid Cystic Carcinoma gene expression profile constructed by ACC-M and ACC-2 at the preliminary stage,we preliminarily screened the referring genes of Notch signal pathway in expression profile on the basis of consulting the related genes of Notch signal pathway by using the methods of bioinformatics. Meanwhile, by using real-time PCR method , we verified the expression level of such genes in ACC-M and ACC-2 cell strains and took the next step to test the obvious differences of the expression in the metastasis and non- metastasis Salivary Adenoid Cystic Carcinoma tissues by the immunohistochemical method. Result: After searching all genes in Notch signal pathway,46 related genes between Notch signal pathway and Salivary Adenoid Cystic Carcinoma were found by analyzing and comparing the gene expression profile through bioinformatics. By using Real-time PCR method to test the expressions of these genes in ACC-M and ACC-2 cell strains,we found that the expressions of RUNX1, Notch-1, Notch-2, Notch-4 in ACC-M were higher than those in ACC-2(P<0.05); the expressions of CHUK, FOXC1, FZD2, FZD3, GBP2 in ACC-M were lower than those in ACC-2(P<0.05) , furthermore, the differences of the expressions of other genes in these two cell strains had no statistical significance(P>0.05). The immunohisto- chemistry result indicated that: the expressions of these genes existed differences in the metastasis and non-metastasis Salivary Adenoid Cystic Carcinoma tissues (P<0.05).This result was the same as that of real-time PCR.Conclusions: Notch signal pathway plays a certain role in the development process of Salivary Adenoid Cystic Carcinoma, the expressions of CHUK, FOXC1, FZD2, FZD3, GBP2 genes may inhibit the metastases of Salivary Adenoid Cystic Carcinoma, while the expressions of RUNX1, Notch-1, Notch-2, Notch-4 genes may boost its metastases. By further research, the foundation is provided by our study to seek for the new promising target in evaluating the progress and recurrence of salivary adenoid cystic carcinoma and the gene treatment.
