ObjectiveTo investigate the effect of CD44 antibody--HI44a,on proliferation and differentiation in HL-60 cells.MethodsExpressions of CD44 transcript of HL-60,NB4,CA46,U266,K562,KAR,CEM,U937 and JURKAT cells were investigated by RT-PCR:Using acute myeloid leukemia cell line HL-60 as the research object,the application of trypan blue staining method,according to HI44a on HL-60 cells,the application of Wright staining and flow cytometry detection HI44a on HL - 60 cells,application of RT-PCR and fluorescence quantitative PCR detection of myeloid cell differentiation-related factor and its receptor signaling pathway(IL-3Rα,GM-CSFRα,Hβc,osteopontin,smad4, TGFβ1,TGFβR1,CD44) gene expression,the application of DNA sequence analysis of full-length osteopontin cDNA.ResultsIn all types of leukemia cell lines,acute promyelocytic leukemia cell line NB4 and acute monocytic leukemia cell line U937 have the highest levels of CD44 mRNA expression;The proliferation of HL-60 cells were inhibited by HI44a;morphology and Detection of cell surface differentiation antigen prompted HL-60 cells to granulocyte change;the expression level of GM-CSFRαand TGFβ1 mRNA were increased,and reduce the level of osteopontin mRNA expression,IL-3Rα,Hβc, TGFβR1,smad4 mRNA expression levels had no significant changes;osteopontin sequence analysis showed that three osteopontin isoforms composition changed. ClnclusionCD44 antibody HI44a can inhibit HL-60 cell proliferation,induction of HL-60 cells to granulocyte differentiation by upregulating GM-CSFRα,TGFβ1 and inhibiting OPN.
